Oncostatin M promotes biphasic tissue factor expression in smooth muscle cells: evidence for Erk-1/2 activation

Abstract: Tissue factor (TF), a transmembrane glycoprotein, initiates the extrinsic coagulation cascade. TF is known to play a major role in mediating thrombosis and thrombotic episodes associated with the progression of atherosclerosis.

“…Indeed, this report is the first to suggest that the scaffolding protein KSR1 may be involved in regulating TF levels by oncogenic EGFR. This is consistent with studies pointing to the involvement of the MEK/Erk1/2 cascade in control of TF expression [21]. However, it should also be borne in mind that the kinase [14,22,23] scaffolding functions [15,24,25] and other functions of KSR1 are still poorly understood and therefore the exact nature of the linkage between this protein and TF remains to be established.…”

“…NF-κB is retained in the cytoplasm associated with a family of inhibitory proteins termed IκB. In response to inflammatory stimuli such as TNF-α and LPS, the IκBs are rapidly phosphorylated, releasing NF-κB and undergoing ubiquitination and proteolysis by the 26S proteosome [33]. In the present study, TNF-α induced translocation of NF-κB (p65) into the nucleus.…”

“…Cells were treated as indicated, followed by exposure to 5nM FVIIa, 150nM FX, and 5mM CaCl 2 in Tris-buffered saline. 20 The chromogenic reaction was triggered by addition of 2mM S-2765 (Chromogenix), stopped with 20 L of 50% acetic acid, read at 405 nm, and the results expressed as arbitrary units normalized to the protein content.…”
