On the Way to Nanometer‐Sized Information of the Bacterial Surface by Tip‐Enhanced Raman Spectroscopy

Abstract: A straight tip: With tip‐enhanced Raman spectroscopy (TERS), chemical and topographical information with nanometric resolution below the diffraction limit of soft biological samples such as bacteria can be gained simultaneously at short measuring times. This noninvasive technique opens new ways for the investigation of many problems in biology and biomedicine.

“…Neugebauer et al measured TERS spectra from Staphylococcus epidermidis [370,371], while Cialla et al measured TERS spectra from tobacco mosaic viruses [372]. Other bacteria-derived samples, such as isolated membrane patches from Halobacterium salinarum have also been measured with TERS, with different patches able to be distinguished [373].…”

“…The applications range from small biomolecules, such as adenine, [25,26] over the biopolymers alginate, [27,28] cytochrome c, [29] and RNA, [30] to living bacterial cells. [31,32] However, all these TERS measurements have been performed in air. Even bacterial cells have been investigated in air.…”

“…Since the first reports in 2000, [1,4,5] TERS has been successfully implemented in different disciplines, such as life science, [6,7] material science, [8] surface science, [9] etc. Besides these applications, TERS has also been used to study some of the most fundamental aspects of its cousin technique, surface-enhanced Raman spectroscopy (SERS), especially the properties of the 'hot' spot in SERS, since TERS is playing with a single 'hot' spot and can collect both the topographic information and the Raman spectrum simultaneously.…”
