On the structure and function of cytochrome b-559

Abstract: A sumary of biochemical, biophysical, and molecular biological data is presented which led to the identification of two different polypeptides (α and β, MW=9.16 and 4.27 kDa) in the cytochrome b-559 protein. The presence of a single His residue on each polypeptide, and the conclusion from spectroscopy that the heme coordination must be bis-histidine led to an obligatory requirement for coordination of a single heme through a heme cross-linked dimer. This structure does not have a precedent among soluble or mem… Show more

“…[21]). Nevertheless, an accumulation of circumstancial evidence suggests a close association of cytochrome b-559 with the oxidizing side of photosystem I1 [l, 5,131, and Cramer et al [7] have very recently proposed a role for this elusive cytochrome as a mediator in the (re)assembly of the watersplitting enzyme in chloroplast development and in response to stress.…”

“…Gel filtration was performed with a Superose 12 HR 10/30 column (Pharmacia) equilibrated with buffer A. An Aminco DW-2 spectrophotometer was used to quantify cytochrome b-559, using the dithionite-reduced minus ferricyanide-oxidized difference spectrum and assuming a differential extiction coefficient of 17 500 M-' cm-' (Cramer et al, 1986).…”
