On the mode of action of intravesical bacillus Calmette-Gurin: in vitro characterization of BCG-activated killer cells

Abstract: Previously we had shown that, upon activation with viable bacillus Calmette-Guérin (BCG), peripheral blood mononuclear cells (PBMNC) could be rendered cytotoxic against otherwise insensitive natural killer (NK)-resistant bladder cancer cell lines. This phenomenon had been termed the BCG-activated killer (BAK) cell phenomenon. By means of depletion and enrichment procedures of mononuclear cell subpopulations derived from BCG-activated PBMNC we further characterized the cytolytic BAK effector cells functionally … Show more

“…238 The mode of action is based on stimulation of the local immune response. 239,240 The immune responses to malignant and virally transformed cells are similar. 241,242 Therefore, topical bacillus Calumette-Guérin treatment has been attempted in the treatment of venereal warts.…”

“…1), the amplification of nonspecific inflammatory response limited to the activation of phagocytes and vascular leakage does not promote the destruction of PDTtreated tumors. In contrast to FIA, BCG treatment triggers a more complex release of immunoregulatory cytokines and other mediators, elicits the engagement of both myeloid and lymphoid immune cells and directly stimulates cell-mediated immunity (24,25,(28)(29)(30).…”

“…However, these observations may reflect solely on the immunological reaction against mycobacterium virulence. The effects implicated in specific antitumoral action of BCG are the activation of macrophages and dendritic cells and associated enhancement of tumor antigen presentation (26,27) as well as the activation of T lymphocytes or lymphoid killer cells (24,(28)(29)(30).…”

“…Further characterization of phenotype BAK-effector cells was consequently the next step in our studies [69, 70]. It is well known that LAK-cells have a maximum cytotoxicity after 2–3 days of stimulation with IL-2 [71].…”
