On the mechanism of Mallory's phosphotungstic acid‐haematoxylin stain

Abstract: The mechanism of Mallory's (1900) phosphotungstic acid-haematoxylin (PTAH) stain is not yet fully understood; staining properties vary with the age of the dye solution. In this study, a 1 year-old solution was employed as described previously. Paper chromatograms revealed blue, red and yellow components of phosphotungstic acid-haematoxylin. Modification of reactive groups as well as consecutive treatment of sections with haematoxylin and phosphotungstic acid indicated hydrogen bond formation by the blue compon… Show more

“…This conclusion was drawn from the differences resulting from the used detection methods. The antibodies bind to actin regardless its quarterly structure [59], which on the other hand is crucial for the phosphotungstic acid hematoxylin staining [60]. Curcumin has been shown to suppress the vascular smooth muscle cells migration and proliferation [61] and to differentially interfere with proteins translation [62], which may correlate with correction of the fibrilar proteins content in the aortic media.…”

“…In this study, Phosphomolybdic Acid (PMA), which targets collagen residues 23 , 24 , was utilized in combination with 1- and 6- micron spatial resolution microCT to assess the postnatal development of mouse lumbar intervertebral discs up to a year of age. Moreover, we specifically report quantitative three-dimensional metrics: the notochord surface area to volume ratio (S.a/V), which defines the structure of the notochord, and the notochord volume.…”

“…In contrast, apart from the aforementioned microorganisms within intestinal lumina, both dipterans showed magenta, coarse to finely granular cytoplasm in ovaries, mosquitoes in hindgut epithelium, and fruit flies in cortical ganglion cells (Figs. [45][46][47]. Similar staining patterns were displayed in salivary gland, cardia/proventriculus, and midgut epithelium of mosquitoes and fruit flies.…”

“…However, PTAH was best at highlighting musculature, as previously reported, 6 given that striated, smooth, and heart musculature were ascertainable. As this dye also binds to Z-bands, 47 cross-striation of muscle fibers was most obvious with PTAH in equine and dipterans, which renders it particularly interesting for the evaluation of myopathies in all species investigated.…”
