On the Determination of Uncertainty and Limit of Detection in Label-Free Biosensors

Abstract: A significant amount of noteworthy articles reviewing different label-free biosensors are being published in the last years. Most of the times, the comparison among the different biosensors is limited by the procedure used of calculating the limit of detection and the measurement uncertainty. This article clarifies and establishes a simple procedure to determine the calibration function and the uncertainty of the concentration measured at any point of the measuring interval of a generic label-free biosensor. “…Most of the FDA approved tests are qRT-PCR, which has lower limit of detection (LoD), hence higher level of sensitivity. Limit of detection (LoD) is one of the crucial performance characteristics that describes the lowest concentration of the target that can be reliably measured by the test ( Lavín et al, 2018 ). This numerical platform can in understanding operational behavior of designed biosensors and reducing their limit of detection.…”

“…The error bars correspond to the standard uncertainty considering the wavelength resolution of the instrument and the standard deviation of n = 100 repeated measurements for each point in the measurements. 35 It can be observed that this uncertainty is about 0.05 nm.…”

“…LoB was determined for each assay using the set of samples without the tested target (Supplementary Table 2 ) and calculated according the formula: LOB = mean blank + 1,645 (SD blank ) 36 . LOD was calculated as: LOD = mean blank + 3,29 (SD blank ) 37 . Specificity of all assays was determined on the sets of samples not containing the target sequence detected by the tested TaqMan probe represented by the genomic DNA isolated from buccal swabs of homozygous females (Supplementary Table 2 ).…”
