Three adult male greyhounds have been repeatedly infected with Toxocara canis by oral administration of 100 to 200 infective ova. Each infection has become patent as shown by the presence of eggs in faeces. Anthelmintic treatment between 7 and 14 weeks after infection yielded an average of 25% of the inoculum as adult worms. These dogs have continued to be fully susceptible to infection despite repeated exposure to this parasite, and despite the presence of scrum antibody to both parasite surface and secreted antigens. These findings imply that adult dogs may not always be immune to toxocaral infection and may therefore contribute significantly to the problem of environmental contamination with T. canis ova.

Zusammenfassung Am Modell der impatent infizierten laktierenden ovariektomierten Hündin wurde der Einfluß verschiedener Oestradiol‐Progesteron‐Kombinationen auf inhibierte Larven am Verlauf der Larvenausscheidung mit der Milch geprüft. Entsprechende Untersuchungen erfolgten an insgesamt 12 Hündinnen. Alle Versuchstiere waren helminthenfrei aufgezogen und einheitlich in den ersten Tagen nach der Geburt einmalig mit 20 000 dritten Larven von Ancylostoma caninum per os oder perkutan infiziert worden. Folgende Ergebnisse wurden erzielt: 1. Durch Applikation von Oestradiol und Progesteron in einem Verhältnis von 1: 20 als Mikrokristallsuspension in einer Dosis von 2,5 mg und 50 mg/Tier konnte bei drei von vier Hündinnen eine Larvenausscheidung mit der Milch induziert werden. Die Zahl der bei diesen Tieren nach der Steroidgabe mit der Milch eliminierten Larven lag jedoch weit unter der, die als Ergebnis der Reaktivierung in der Folge einer Gravidität während einer 2. Laktation p. i. zu erwarten gewesen wäre. 2. Der Versuch, die doppelte und dreifache Dosis der Steroide an den gleichen Tieren 4 und 8 Wochen nach der ersten Stcroidgabe zu prüfen, gelang nur bei zwei von vier Hündinnen. Bei einem dieser Tiere war die Larvenausscheidung nach der doppelten Dosis 8mal, nach der dreifachen Dosis 16mal größer als nach der Initialdosis. Die Dosisabhängigkeit der Steroidwirkung wurde bei der zweiten Hündin infolge zu geringer impatenter Infektion nicht deutlich. 3. Nach Applikation von Oestradiol und Progesteron in einem Verhältnis von 1: 40 in öliger Lösung bei täglich um 0,01 mg und 0,4 mg steigender Dosis von 0,08 mg und 3,2 mg bis 0,21 mg Oestradiol und 8,4 mg Progesteron am 14. Tag der Behandlung konnte während und nach der Steroidgabe eine Larvenausscheidung mit der Milch induziert werden, die in Umfang und Ablauf weitgehend der entsprach, die als Ergebnis einer Reaktivierung wahrend der Graviditát in einer 2. Laktation p. i. zu erwarten gewesen wäre. In allen Fällen, in denen eine Larvenausscheidung mit der Milch durch Oestradiol‐Progesteron‐Applikation zu induzieren war, bestanden zwischen der Larvenausscheidung mit der Milch und der oestrogenen Wirkung auf das Tier, gemessen am Eosinophilie‐ und Pyknose‐Index des Scheidenepithels, signifikante Korrelationen. Die Untersuchungen erfolgten im Rahmen des Sonderforschungsbereiches 146 „Versuchstierforschung” der Deutschen Forschungsgemeinschaft. Aim of the investigation was to examine whether two administrations of moxidectin to pregnant dogs could prevent pre-natal and lactogenic infections of puppies with reactivated Toxocara canis larvae. Four pregnant beagles, infected experimentally with 20 000 embryonated eggs of T. canis, were treated subcutaneously with 1 mg moxidectin per kg body weight on days 40 and 55 of pregnancy (5-13 days before parturition). One further dam and its puppies served as untreated control. Two applications of moxidectin completely prevented pre-natal and lactogenic infections in the puppies. Neither intestinal stages nor somatic larvae were found in the dams or their corresponding puppies. All puppies and dams of the treatment group remained coproscopically negative until 42 days after parturition. The administration of moxidectin did not show any side effects in the dams. None of the puppies of the treated dams showed any pathological abnormalities. In the untreated dam one adult and 26 somatic larvae of T. canis were detected at necropsy. All puppies of the untreated dam showed a patent T. canis infection from day 28 post-natum (p.n.); 296 pre-adult and adult stages of T. canis were spontaneously eliminated and 51 intestinal stages and five somatic larvae of T. canis were recovered at necropsy. In contrast to the puppies of the treated dams all negative control puppies showed blood eosinophilia after parturition and elevated liver enzyme levels.
