On‐Off PVC Membrane Based Potentiometric Immunosensor for Label‐Free Detection of Alpha‐Fetoprotein

Abstract: A poly(vinylchloride) (PVC) membrane based potentiometric immunosensor for the direct detection of alphafetoprotein (AFP) has been developed. First, Au colloid particle was chemisorbed upon amino groups of ophenylenediamine, which were dissolved in plasticized PVC membrane. Then alpha-fetoprotein antibody (anti-AFP) was immobilized upon the surface of the Au colloid particle to prepare a potentiometric AFP immunosensor. The Au colloid particle modified PVC membrane was characterized by digital photo and transm… Show more

“…Reusing of an immunosensor is very appealing due the possibility of application to many samples in a short period at a low cost relatively to the disposable ones [54]. Accordingly, several regeneration reagents which have already been successfully used by Park et al [55].in 2000 were tested.…”

“…The pH value of the detection solution was also an important parameter, and unsuitable pH may cause protein denaturalization or immunosensor unstable. 38 As shown in Fig. 5B, the peak current change was increased with the increment of pH value from 5 to 7 and then decreased.…”

“…The membrane composition was as follows: 3.0 wt% o-phenylenediamine, 29.8 wt% membrane solvent DOP and 67.2 wt% PVC. The o-phenylenediaminemodified PVC membrane was fixed on the easy-replace plastic part and joint with PVC tube [27]. After washing with distilled water, the aminated PVC membrane electrode was dipped into 16-nm gold colloids for 4 h at 4 8C.…”
