Oligosaccharides obtained by partial hydrolysis of lipopolysaccharide from Fusobacterium nucleatum F1

Abstract: Lipopolysaccharides isolated from Fusobacterium nucleatum (F. nucleatum) strain Fl was split by hydrolysis with 1% acetic acid into water‐soluble polysaccharide and insoluble material. Gel filtration of the water‐soluble polysaccharide on Bio‐Gel P‐60 gave a serologically active fraction eluted at the void volume (Vo) of the column with a sugar composition similar to that of the parent LPS and a fraction eluted at 2.5 × Vo. The latter, supposed to represent the core polysaccharide, contained phosphorus, glucos… Show more

“…LPS from oral strains of F. nucleatum consist of a typical lipid A component, exhibiting a close structural relationship to that of other groups of gram-negative bacteria (123), and an O-antigen heteropolysaccharide (133,174) and resemble LPS of other gram-negative bacteria (60). The LPS of F. nucleatum contain 3-deoxy-D-manno-octulosonic acid (KDO) (84,131,132). F. nucleatum strains have been classified into six chemotypes on the basis of the polysaccharide composition of their LPS (84).…”

“…The amount of preparations of other bacterial strains separated on SDS-PAGE were visually adjusted after Coomassie brilliant blue staining according to the amount of the F. nucleatum Fevl sample. The extraction of LPS from F. nucleatum Fevl whole cells with phenol [23] and treatment with pronase, DNAase and RNAase was as described [24]. This preparation contained less than 1% amino acids with reference to the amount of glucosamine, as judged from amino-acid analysis of acid hydrolysates [22].…”
