Oligonucleotide microarray analysis of ameloblastoma compared with dentigerous cyst

Abstract: We identified important genes related to the development and progression of ameloblastoma through a large-scale gene expression analysis. This study will stimulate further investigations on genes significant for early diagnosis and prognosis of ameloblastoma.

“…This study focused on the description of the gene expression of malignant odontogenic tumors; we did not find any of those genes significantly expressed in our samples. Lim et al, in 2006, reported an oligonucleotide microarray analysis of 2 ameloblastomas compared with a dentigerous cyst (Lim et al, 2006). Overexpressed genes postulated to play a role in the tumorigenesis of ameloblastoma included VAV3, EGFR, TOM1L1, FUBP1, CDK2, TNKS, PTHRP, SMARCA2, KLK5, and CLU.…”

“…There have been two microarray studies of ameloblastoma (Heikinheimo et al, 2002;Carinci et al, 2004), one of them using cDNA microarrays containing 19,000 human cDNAs, and, more recently, an oligonucleotide analysis of whole-tissue ameloblastoma in a 34,000 human RNA microarray (Lim et al, 2006). The objectives of our study were: (a) to characterize the gene expression profile of laser-captured microdissected ameloblastoma cells using oligonucleotide microarray technology; (b) to compare the gene expression profile with universal human reference RNA; and (c) to identify genes or gene products that may have diagnostic, prognostic, or therapeutic potential.…”

“…Therefore, it is necessary to further elucidate other effects of COX-2 in the epithelium and connective tissue of odontogenic lesions and to compare dentigerous cyst with ameloblastoma. Previous studies have shown that each of ameloblastoma and dentigerous cyst presents a distinct evolution and biological behavior, and in addition to their clinical and behavioral differences (57), they reflect diversities in various features such as gene expression (58), pattern of expression of cytokines (3), interaction of different proteins, and collagen components (57).…”

“…Expression of RB, the most important cell cycle regulator, has been lower in ameloblastomas than in dentigerous cysts, but higher in ameloblastic tumors than in tooth germs (58)(59). Cyclin D1 is strongly implicated in cell cycle progression by regulating the transition from G1 to S phase, while CKIs, p16…”
