Oleuropein alleviates gestational diabetes mellitus by activating AMPK signaling

Abstract: Background: Gestational diabetes mellitus (GDM) has a high incidence rate among pregnant women. The objective of the study was to assess the effect of plant-derived oleuropein in attenuating inflammatory and oxidative stress of GDM. Methods: Oleuropein was administered to GDM mice at the doses of 5 or 10 mg/kg/day. Body weight, blood glucose, insulin and hepatic glycogen levels were recorded. To evaluate the effect of oleuropein in reducing oxidative stress, enzyme-linked immunosorbent assay (ELISA) was used … Show more

“…Oleuropein (OP), which exists in olive leaves, unripe olive fruits, olive branches, and olive oil, is a highly bioactive phenolic compound with various biological functions [ 7 , 8 ]. Previous studies have shown that OP consumption exerts therapeutic effects on the early stage of type 2 diabetes by repairing islet morphology, regulating insulin secretion, and improving glucose tolerance and insulin sensitivity [ 9 , 10 , 11 , 12 ]. However, the alleviating effects of OP on the advanced stage of type 2 diabetes and the regulatory effects of OP on intestinal microbes have not yet been investigated.…”

“…Oleuropein treatment with the dose of 5 and 10 mg/kg decreases blood glucose, insulin, and hepatic glycogen and improves glucose tolerance, insulin resistance, and islet structure in gestational diabetes mellitus mice. These results are associated with AMPK signaling activation in the liver …”

“…IL-1β promoted secretion of IL-8 and IL-6 and induced apoptosis of islet β cells, and IL-8 and IL-6 stimulated infiltration of inflammatory cells to aggravate the inflammatory injury [3]. Suppression of IL-6, IL-1β, and TNF-α ameliorated GDM [11]. Deletion of IRF2BP2 exacerbated high-fat diet-induced increases in IL-1β and TNF-α in mice [8].…”
