“…It was found to readily degrade, with a visible colour change in aqueous solutions from yellow to blue [65]. During pharmacokinetic studies using rats, it was found that the acidic olanzapine solutions that were being continuously delivered to the rats were degrading, resulting in a colour change from bright orange to dark green, with formation of an unknown precipitate [66].…”

“…An alternative method of administration involves an embedded infusion pump, and several studies have reported use of an osmotic pump (Chintoh et al, 2008;Choi et al, 2007;van der Zwaal et al, 2008). However, no study using this implanted osmotic pump has noted antipsychotic-induced metabolic side effects such as weight gain or adiposity in male rats.…”

“…One reason for this difficulty is that olanzapine is chemically unstable in acidic solution, which was frequently used to dissolve olanzapine in previous studies. Indeed, plasma concentration of olanzapine was found to decrease to 10% after 4-week infusion with osmotic pump (van der Zwaal et al, 2008).…”

“…Pure drug samples of olanzapine (2-methyl-4-(4-methyl-1-piperazinyl)-10H thieno [2,3][1,5]benzodiazepine) and flouxetine were kindly provided by Miracle Pharmaceuticals (Islamabad, Pakistan). HPLC-grade acetonitrile, methanol and ethanol were obtained from Merck (Germany) and used without further purification or filtration.…”

“…Olanzapine has shown to have affinity with dopamine D1 and D2, muscarinic and serotonin 5HT2 receptors [3]. Plasma concentration of olanzapine has a correlation with its clinical effects and also shown to have individual variability in its pharmacokinetics [4,5].…”
