OK-432 Induces Production of Neutrophil Chemotactic Factors in Malignant Pleural Effusion.

Abstract: Weinvestigated the changes in cellular components and neutrophil chemotactic factors in pleural fluid from 19 lung cancer patients who received intrapleural injection of OK-432 to treat malignant pleurisy. Not only neutrophil chemotactic activity (NCA) but also neutrophil count and percentage were increased significantly at 6 hours after OK-432injection. The neutrophil count was significantly correlated with NCAlevel. The levels of C5a and IL-8 in pleural fluid were increased significantly after OK-432 injecti… Show more

“…It is crucial to know whether these regressions of MPE are mediated by specific (CTL) or aspecific (NK) antitumoral effector cells, or only by the sclerosing effect secondary to the release of inflammatory cytokines, such IL-8, by the mesothelial cells. Thus, it was clearly demonstrated that the control of MPE by instillation of OK-432 tetracycline, Corynebacterium parvum was not mediated by immune cells, but by the inflammatory response induced by the release from mesothelial cells of C5a, IL-8, IL-6 or TNF, leading to pleural sclerosis [177,179,180].…”

“…40 Although the chemotactic activity and vascular permeability-increasing activity of C5a are thought to increase the adhesion of neutrophils to the peritoneal vascular network, 41 we focused our attention on milky spots. The milky spots of the greater omentum are important tissue for intraperitoneal immunity, involved in the migration of neutrophils, macrophages, and lymphocytes.…”

“…In rats, OK-432 (0.3 KE kg −1 ) is less effective than talc (400 mg kg −1 ) in inducing pleurodesis at 30 days [33]. In Japan, it is regarded by some as the sclerosing agent of choice [34]. Response as high as 75 % has been achieved.…”
