Targeted capture of nuclear genes increasingly contributes to unravelling phylogenetic relationships that hitherto remained unresolved because of limitations of traditional Sanger sequencing. In particular, the study of tropical plant families has been compromised because they often rely on highly degraded DNA obtained from herbarium specimens. One such example is the pantropical Ochnaceae, which comprises 33 genera and approximately 550 species, occurring mostly in savannas and moist tropical forests. Here, we developed a set of baits covering about 660,000 bp from 275 nuclear genes used for the targeted enrichment based on all but one genus and more than 250 species. Using this novel dataset, we resolved the phylogenetic backbone of Ochnaceae, including that of Ochninae, and we established new relationships. Most importantly, our findings highlight that the neotropical and palaeotropical taxa of Sauvagesia form independent clades, requiring the re-erection of formerly separate genera. Ouratea and Ochna (both Ochninae), by far the most species-rich genera and represented by 120 and 41 species, respectively, came out as monophyletic. In contrast, the third-most species-rich genus, Campylospermum, is polyphyletic in two distinct clades. Ouratea, the only neotropical genus of Ochninae, was sister to the five palaeotropical genera of this subtribe. The bait kit developed in this study proved to be particularly useful for unravelling relationships within Ochninae, which includes about two-thirds of the species diversity in the family. Buds usually possess mechanical or chemical protection and may also have secretory structures. We discovered an intricate secretory system in Ouratea castaneifolia (Ochnaceae) related to the protection of buds and young leaves. We studied this system, focusing on the distribution, morphology, histochemistry, and ultrastructure of glands during sprouting. Samples of buds and leaves were processed following the usual procedures for light and electron microscopy. Overlapping bud scales protect dormant buds, and each young leaf is covered with a pair of stipules. Stipules and scales possess a resin gland, while the former also possess an extrafloral nectary. Despite their distinct secretions, these glands are similar and comprise secreting palisade epidermis. Young leaves also possess marginal colleters. All the studied glands shared some structural traits, including palisade secretory epidermis and the absence of stomata. Secretory activity is carried out by epidermal cells. Functionally, the activity of these glands is synchronous with the young and vulnerable stage of vegetative organs. This is the first report of colleters and resin glands for O. castaneifolia. We found evidence that these glands are correlated with protection against herbivores and/or abiotic agents during a developmental stage that precedes the establishment of mechanical defenses.

Rhytidanthera is the only genus of Ochnaceae with compound leaves, and it forms sparse, small populations restricted to the primary forest of the Colombian and Venezuelan Andes and the sandstone hills of La Macarena and Chiribiquete. It has been suggested to be related to Godoya, Cespedesia and Krukoviella in taxonomic and phylogenetic studies. However, the relationships in this group remain poorly resolved, due to the lack of a wider molecular and taxonomic sampling. We present a phylogenetic study of tribe Sauvagesieae including three Rhytidanthera spp. and one species each of Godoya and Cespedesia, using sequences from one nuclear (ITS) and four plastid DNA regions (matK, ndhF, rbcL and trnL-F), in order to clarify the relationships among these four genera. We recover a new and well-supported phylogenetic hypothesis, with Rhytidanthera as sister to a clade comprising Godoya and the clade formed by Cespedesia and Krukoviella. This Neotropical clade was recovered as sister to the monotypic and African genus Fleurydora. The clade comprising Rhytidanthera, Godoya, Cespedesia and Krukoviella is supported by the presence of glandular structures (colleters) at the base of stipules and bracts. The colleters also evolved at the base of the sepals in this clade, with a secondary loss in Cespedesia and Krukoviella. We suggest that the combination of unequal-sized sepals enclosing the floral bud and the presence of colleters in members of this clade could have a role in the protection of leaves and flowers in the early stages of development, instead of acting in pollinator attraction.
