In 2012, a pink mold rot was observed on apple (Malus pumila var. dulcissima Koidz.) at the Wholesale Market for Agricultural Products, Jinju, Korea. The first symptom of pink mold rot on apple fruit is a water-soaked appearance of the affected tissue and surface fruit formed pink mold rot, then became brown and produced a mass of powdery pink conidia spores. Colony was fast growing colonies, pinksh, zonate in diurnal rhythm, powdery from conidia. Optimum temperature for mycelial growth was 25 o C. Conidia were hyaline, smooth, two-celled, thick-walled conidia with truncate bases, ellipsoidal to pyriform, and characteristically held together zig-zag chains and 12-26 × 8-12 µm in size. Conidiophore was erect, colorless, unbranched, and 4-5 µm wide. On the basis of mycological characteristics, pathogenicity test, and molecular identification with the ITS region, the causal fungus was identified as Trichothecium roseum (Pers.) Link ex Gray.

-Pink Rot on melon and White Stain Symptom on grape are caused by Trichothecium roseum, one of the most important diseases of grape and melon. These diseases have been occurred in national-wide in Korea and causes irreversible damage on the grape and the melon at harvest season. This research presents the evaluation of the capacity of Bacillus subtillis HK2 to protect both melon and grape against T. reseum and establishes its role as a biocontrol agent. In this study, we isolated a Bacillus strain HK2 from rhizosphere soil, identified it as Bacillus subtillis by 16S rRNA analysis and demonstrated its antifungal activity against T. roseum. Under I-plate assay it was observed that the effect of hyphal growth inhibition was not due to production of volatile compounds. The optimum culture condition of HK2 was found at 30℃ and initial pH of 7.0. Application of HK2 culture suspension reduced 90.2% of white stain symptom on grape as compared to control, resulting in greater protection to grape against T. roseum infestation. Butanol extract of HK2 culture purified using flash column chromatography. The antifungal material was a polar substance as it showed antifungal activity in polar elute. Therefore, our results indicated a clear potential of B. subtilis HK2 to be used for biocontrol of Pink rot in melon and white stain symptom on grape caused by T. roseum.

During 2017–2019, we observed Trichothecium spp. causing fruit rot in the field and in storage. This study was conducted to examine morphological differences of the species from different hosts, reveal the species’ potential host range, and evaluate the efficacy of five fungicides. Six strains of Trichothecium spp. isolated from nectarine, peach and walnut were selected. Although the colony morphology, mycelial growth rate and spore size differed among hosts, phylogenetic analysis based on the internal transcribed spacer and part of the 5ʹ end of the β‐tubulin gene showed that all tested strains belonged to Trichothecium roseum. For its host range, 23 kinds of fruit were examined using T. roseum strain YT‐1 as an inoculum; 10 kinds of fruit, including pear, apple, mango, Chinese chestnut, pepino melon, fig and durian, were susceptible to T. roseum, with minimum inoculation concentrations ranging from 104 to 105 spores/ml. The fungicides that most effectively inhibited the six isolates were fluazinam and fludioxonil, with EC50 values of 0.07–0.1 and 0.01–0.04 μg/ml, respectively, followed by difenoconazole (0.81–2.96 μg/ml), boscalid (5.43–13.51 μg/ml) and azoxystrobin (9.18–27.25 μg/ml). Improvement of the shelf life of nectarines held in plastic trays was explored using allyl isothiocyanate (AITC) against T. roseum YT‐1. The application of 10 μl/L AITC significantly suppressed disease symptoms. The findings provide useful information for future disease emergency management in the field and for food preservation.
