Cyanobacteria, which develop abundantly in aquatic ecosystems, can be harmful to humans and animals not only by releasing toxins that cause poisoning but also by provoking cytogenetic effects. The influence of the mass development of cyanobacteria on the genotoxic properties of natural water has been studied in model ecosystems (microcosms) with different compositions of biotic components (zooplankton, amphipods and fish). The validated plant test system “Allium test” was used in this study. Genotoxic effects were detected at microcystin concentrations below those established by the World Health Organization (WHO) for drinking water. In all experimental treatments, cells with disorders such as polyploidy and mitotic abnormalities associated with damage to the mitotic spindle, including c-mitosis, as well as lagging chromosomes were found. Genotoxic effects were associated with the abundance of cyanobacteria, which, in turn, depended on the composition of aquatic organisms in the experimental ecosystem. Fish, to a greater extent than other aquatic animals, maintain an abundance of cyanobacteria. After one month, in microcosms with fish, mitotic abnormalities and polyploidy continued to be detected, whereas in other treatments, there were no statistically significant genotoxic effects. In microcosms with amphipods, the number and biomass of cyanobacteria decreased to the greatest extent, and only one parameter of genotoxic activity (frequency of polyploidy) significantly differed from the control.

Background Excess loads of nutrients finding their way into waterbodies can cause rapid and excessive growth of phytoplankton species and lead to the formation of cyanobacterial harmful algal blooms (cyano-HABs). Toxic cyanobacteria produce a broad range of bioactive metabolites, some of which are known as cyanotoxins. These metabolites can negatively impact the ecosystem, and human and animal health, thus their presence needs to be closely monitored and mitigated. This study aimed to monitor St. George Lake (Athalassa National Forest Park, Cyprus) for its water quality characteristics, and initiate a new methodology to control the bloom that occurred in the lake during summer 2019, by comparing hydrogen peroxide treatment with novel metallic peroxide granules as source of hydrogen peroxide. Results Lake monitoring showed that pH, salinity, total dissolved solids and conductivity varied throughout the year, and nutrients concentration was high, indicating a eutrophic lake. The cyanobacterium Merismopedia sp. bloomed in the lake between June and September 2019, comprising up to 99% of the phytoplankton biovolume. The presence of microcystin synthase encoding gene (mcyB, mcyE) was documented, however microcystins were not detected by tandem mass spectroscopy. Treatment with liquid hydrogen peroxide in concentrations 1 to 5 mg L−1 had no effect on the phycocyanin fluorescence (Ft) and quantum yield of PSII (Fv/Fm) indicating an ineffective treatment for the dense Merismopedia bloom (1 million cells mL−1 ± 20%). Metallic peroxide granules tested for their H2O2 releasing capacity in St. George Lake water, showing that CaO2 released higher H2O2 concentration and therefore have better mitigation efficiency than MgO2 granules. Conclusion The present study highlights the importance of monitoring several water parameters to conclude on the different actions to be taken to limit eutrophication in the catchment area. The findings demonstrated that testing for the presence of genes involved in cyanotoxin production may not be sufficient to follow cyanotoxins in the water, therefore it should be accompanied with analytical confirmation. Treatment experiments indicated that slow release of H2O2 from peroxide granules may be an alternative to liquid hydrogen peroxide when applied in appropriate doses, but further investigation is needed before it is applied at the field. The aim of the work was to investigate the effect of cyanobacteria and elodea on population characteristics (abundance, biomass, fecundity) and “health” indicators (embryo status, heart rate and thermal resistance) of aquatic invertebrates in microcosms with controlled conditions. In July-August 2018, four treatments with environmental conditions of the coastal zone of the Rybinsk Reservoir during the period of cyanobacterial bloom were modeled. Mass species of plankton (with 80% dominance of large Daphnia longispina) and benthic animals (Unio pictorum, Gmelinoides fasciatus, Asellus aquaticus) and Elodea canadensis were placed in microcosms. In treatments with cyanobacteria, the toxic microcystins-LR were detected (24-53% of the sum of toxins, or 6,6-66,5 µg/l). The cyanobacteria Aphanizomenon flos-aquae, Microcystis aeruginosa and Gloeoitrichia spp. affected the structure of plankton reflecting in a decrease in the amount of green algae and an increase in the abundance of small crustaceans. Under the influence of cyanobacteria, variables of benthic animals differed significantly from control treatment, including a reduced level of thermal resistance of mollusks and amphipods, an increased recovery time of the heart rate after loading (salinity test) in mollusks, a decrease in individual fecundity, and an increase in the frequency of malformations and embryo mortality in amphipods. The presence of elodea enhanced these effects. Thus, the mass development of cyanobacteria and elodea affects the physiological status of sensitive species of aquatic animals, reducing their adaptability to natural stresses and worsening the quality of offspring.
