Occurrence of Gluconacetobacter diazotrophicus in tropical and subtropical plants of Western Ghats, India

Abstract: Endophytic bacteria were isolated from the tissues of surface sterilized roots, stems, and leaves of fifty different crop plants. Phenotypic, biochemical tests and species-specific PCR assay permitted identification of four isolates of Gluconacetobacter diazotrophicus from root tissues of carrot (Daucus carota L.), raddish (Raphanus sativus L.), beetroot (Beta vulgaris L.) and coffee (Coffea arabica L.). Further the plant growth promoting traits such as nitrogenase activity, production of phytohormone indole a… Show more

“…The samples were macerated in 1% sucrose solution using a low-speed blender for 30 s. The homogeneous mixture obtained was serially diluted from 10 -1 to 10 -3 ; then, 100 μL aliquots were plated in triplicate in 6-mL vials containing 3 mL of semisolid LGI-P medium (Cavalcante and Döbereiner, 1988;Madhaiyan et al, 2004).…”

“…For this reaction, the universal primers RB and RM (Madhaiyan et al, 2004) were used; the expected fragment was approximately 800 bp. The final volume of the PCR reaction was adjusted to 25 µL (1X PCR buffer, 0.2 mM dNTPs, 1 mM MgCl2, 0.5 μM primers, 1 U of polymerase taq DNA recombinant polymerase by Invitrogen and 100 ng of bacterial DNA).…”

“…Among the different microorganisms used in sweet corn cultivation, the nitrogen fixing microorganisms like Acetobacter, phosphorous solubilizing bacteria and potassium solubilizing bacteria are used extensively in single, double and triple inoculant formulation however the native efficient isolates are not concentrated much in sweet corn production. Madhaiyan et al, (2004) Conceptual design is important in developing new technologies and also utilization of different beneficial plant growth promoting rhizomicroorganisms for sustainable vegetable production. The basis of conceptual design is simply to first convince a model and then to devise a strategy and method for achieving the reality.…”
