Occurrence of a novel mastrevirus in sugarcane germplasm collections in Florida, Guadeloupe and Réunion

Abstract: BackgroundIn Africa and Asia, sugarcane is the host of at least seven different virus species in the genus Mastrevirus of the family Geminiviridae. However, with the exception of Sugarcane white streak virus in Barbados, no other sugarcane-infecting mastrevirus has been reported in the New World. Conservation and exchange of sugarcane germplasm using stalk cuttings facilitates the spread of sugarcane-infecting viruses.MethodsA virion-associated nucleic acids (VANA)-based metagenomics approach was used to detec… Show more

“…A BLASTn search against the GenBank nr database using a 3137 base contig from maize sample #76 (Supplementary Table 1 ) identified a 168-nucleotide region with ~ 81% identity to the Rep coding sequence (CDS) of sugarcane striate virus (SCStV), a Mastrevirus [ 51 ]. Subsequent evaluation of this contig by a BLASTx search revealed potential homology (~ 31% query coverage with 39–44% amino acid identity) to the RepA proteins of SCStV and maize streak virus (MSV), which is the type member of the Mastrevirus genus.…”

“…As a proof of concept, this new metagenomic methodology was applied to a set of non-cultivated plants sampled from a single farm on Reunion Island. In addition to finding viruses that belong to two of the six mastreviruses species that have previously been detected on Reunion Island, MSV and maize streak Reunion virus (MSRV) 6,17 , this article further highlights the gaps that likely remain in our appreciation of mastrevirus diversity on this island by discovering mastreviruses that could represent three new mastrevirus species.…”

“…At the present time, the best methodology for virome identification in a specific environment is viral metagenomics, and different approaches are available [ 21 , 22 ]. Virion-associated nucleic acid (VANA) metagenomics has already been used to identify known and new viruses in sugarcane [ 5 , 7 , 23 ] and in wild Poaceae [ 24 , 25 , 26 ]. The objective of this study was to use this methodology for the characterization and comparison of the virome of sugarcane varieties located at the quarantine glasshouse of Visacane and the virome of wild Poaceae present in the surrounding environment of this sugarcane quarantine station.…”
