Observations on the Isolated Phrenic Nerve Diaphragm Preparation of the Rat

Abstract: The work which led up to the investigation described in this paper was concerned with the influence of adrenaline on the action of acetylcholine or on tissue functions normally elicited by acetylcholine. Previously experiments were carried out on animals with a normal circulation or on perfused organs, and any adrenaline effects observed were accompanied by vasoconstriction, so that it was conceivable that its action was due to vascular changes. The improvement in the transmission of impulses along the motor ne… Show more

“…The mice were anaesthetized with isoflurane (Cristalia®, Itapira, SP, Brazil), via inhalation and later exsanguinated by sectioning and bleeding the cervical vessels. The phrenic nerve-diaphragm preparation was removed according to the protocol proposed by Bülbring and Bowman (1997) adapted for mice and placed in a 5 mL organ bath containing Tyrode solution, composition (in mM): NaCl 137; KCl 2.7; CaCl2 1.8; MgCl2 0.49; NaH2PO4 0.42; NaHCO3 11.9, and Glucose 11.1, pH 7.0. Preparations were maintained at 37 °C and aerated with carbogen (95% O2/ 5% CO2).…”

“…The left side of the diaphragm with the phrenic nerve was dissected from the rat following the procedures described previously (Bulbring 1997). The phrenic nerve diaphragm was kept in the Tyrode's solution with 95% O 2 and 5% CO 2 .…”

“…The phrenic nerve-diaphragm was removed from anesthetized mice (chloral hydrate, 3 mg/kg), which were euthanized via exsanguination. The left diaphragm was removed as previously described [50]. The toxin preparations were suspended and maintained as previously described [51].…”
