Aim:The aim was to determine the motion and kinematics characteristic of frozen-thawed spermatozoa in Sirohi goat using computer-assisted semen analysis. Materials and Methods:A study was carried out in Sirohi buck. Semen collection was made biweekly from each buck with the help of artificial vagina. A total of 12 ejaculates were collected from two bucks (six ejaculates from each buck). Freshly collected semen was pooled and later evaluated. The pooled semen sample was extended with standard glycerolated egg yolk tris extender and later subjected to a process of cryopreservation. The motion and kinematic characteristics of spermatozoa were studied during freez-thawing process.Results: Significantly (p<0.01) higher value of live percent, hypo-osmotic swelling test, and acrosomal integrity were recorded in neat semen followed by diluted and frozen thaw semen. The proportion of spermatozoa showing slow progression were the highest in the neat and diluted semen followed by rapid and non-progressively motile, while a reverse pattern was observed in the frozen thaw semen where the proportion of non-progressively motile spermatozoa were significantly (p<0.01) higher followed by slow and rapid progression. Conclusion:This study showed that the best results for motion, vitality, plasma membrane integrity, and acrosome status were obtained in the neat semen followed by diluted and frozen thaw semen. Further, the process of cryopreservation results in a shift of motility from slow to non-progressive in the post-thaw semen with a significant decrease in the path velocities when compared to neat and diluted semen. Hence, it can be concluded that freezing-thawing process reduces the motility and kinematic characters spermatozoa and may be an important factor affecting the fertilizing ability of spermatozoa resulting in poor conception rate after insemination in goats.

This study aimed to evaluate various concentrations of egg yolk (5, 10, or 20%) in combination with different concentrations of glycerol (3% or 6%) added to a Tris-based extender on the post-thaw characteristics of sperm obtained from Tayassu tajacu. For this purpose, semen from 10 sexually male mature collared peccaries was collected by electroejaculation and evaluated for sperm motility, vigour, viability, morphology and functional membrane integrity. The ejaculates were initially extended in Tris-fructose plus egg yolk (5%, 10% or 20%). After cooling, the semen was added to Tris-egg yolk plus glycerol (6% or 12%), resulting in a final concentration of 3% or 6% glycerol of the extender. Straws were frozen using liquid nitrogen and thawed in a water bath at 37°C for 30 s. The frozen-thawed semen was evaluated as reported for fresh semen. After thawing, a significant decrease was verified for sperm motility and vigour, for all the samples in comparison with fresh semen. However, no differences were evidenced among treatments for any sperm characteristics evaluated (p > 0.05), except for the combination between 10% egg yolk and 6% glycerol, which provided the worst preservation of functional membrane integrity (p < 0.05). The interactions between higher concentrations of egg yolk (20%) and glycerol (6%) and also between lower concentrations of the same substances (5% egg yolk and 3% glycerol) added to the Tris-based extender negatively affected the preservation of the normal sperm morphology after thawing (p < 0.05). In conclusion, the use of Tris-based extender added to 10% or 20% egg yolk plus 3% glycerol is recommended for effective sperm cryopreservation in collared peccaries.

Resumo O objetivo do presente estudo foi testar a dimetilacetamida (DMA) em diferentes concentrações, associada ou não ao glicerol (GL), sobre a viabilidade espermática do sêmen ovino congelado. Foram utilizados 10 ejaculados de dois carneiros adultos da raça Santa Inês. Os ejaculados foram divididos em sete grupos experimentais, respeitando o limite máximo de 5% de DMA, sendo eles: GL6%, DMA3%, GL5%+DMA1%, GL4%+DMA2%, GL3%+DMA3%, GL2%+DMA4%, GL1%+DMA5%. Os espermatozoides criopreservados nos diferentes tratamentos foram analisados quanto à cinética subjetiva, integridade estrutural da membrana plasmática (EOS), integridade funcional da membrana plasmática (CO) e morfologia espermática, observando defeitos totais (DT) e defeitos maiores (DM). A motilidade total (MT) e a progressiva (MP) pós-descongelação nos grupos GL5%+DMA1%; GL4%+DMA2% e GL3%+DMA3%, foram semelhantes (P>0,05) ao tratamento controle (GL6%). Destes, o diluidor GL4%+DMA2% foi o único que promoveu a manutenção da MT e MP pós-descongelação, quando comparado com o sêmen in natura (P>0,05). Não foram observadas diferenças significativas (P>0,05) para os parâmetros de EOS, CO, DT e DM nos diferentes grupos avaliados. A dimetilacetamida associada ao glicerol mostrou-se eficaz na manutenção da viabilidade espermática em ovinos, avaliada pós-descongelação. Entretanto, foi observado efeito deletério da DMA nas concentrações mais elevadas ou quando não esteve associada ao glicerol.
