Objective analysis of cancer stem cell marker expression using immunohistochemistry

Abstract: Analysis of immunohistochemical expression is often a subjective and semiquantitative process that can lead to the inconsistent reporting of results. To assess the effect that region selection and quantification method have on results, five different cancer stem cell markers were used in this study to compare tissue scoring with digital analysis methods that used three different tissue annotation methods. Samples of tumour and normal mucosa were used from 10 consecutive stage II colon cancer patients and stain… Show more

“…One of the difficulties of research on stem cell markers is that the evaluation method varies widely for different studies and the results obtained can be inconsistent. 11 We used 5% as a cut-off level for LGR5 based on a previous study on colorectal cancer using the same antibody. 13 In order to ensure maximum credibility of the results, we used pathological sections which included not only the invasive front but also the normal colon tissue whenever possible, so that the normal part can be used as an internal control.…”

“…Multiple pathologists should score a set of cases to investigate how well the new scoring method is reproducible. Digital imaging analysis showed good results in liver fibrosis [ 25 ], in assessing digital ulcers in patients with systemic sclerosis [ 1 ], and in analysis of cancer stem cell marker expression [ 20 ]. This type of tissue analysis might be a good alternative for scoring of MoM periprosthetic tissue.…”

“…Sections were scanned on a high-resolution scanner (Aperio Scanscope XT; Leica Biosystems, North Ryde, New South Wales, Australia) and manually annotated to ensure the tissue analysed matched the histology type to which samples had been assigned. Image analysis software was then used to quantify marker expression as previously described (Aperio Imagescope version 11) ( 35 , 36 ). Expression of PD-L1 by immune and tumour cells was included in the analysis.…”
