The neuronal cells are distributed into discrete histological layers in the fully differentiated retina ( Fig. 1 ), with the ganglion cell layer arranged proximal to the vitreous and the photoreceptor cells (rods and cones) residing at the opposite side of the neural retina, just underneath the retinal pigment epithelium (RPE) layer. The RPE is a monolayer of polarized epithelial cells that serves as the interface between the neural retina and the choroid, the blood supply to the outer cell layers of the retina. The apical tips of the photoreceptor outer segments (OS) are adjacent to and invaginate into the apical face of the RPE (one of the few examples of apical-apical cellular contact in the body) ( Fig. 1C ). Bruch's membrane (BrM; Fig. 1B, C ) is not a membrane, per se, but rather a pentalaminar structure that consists of the basement membranes of the RPE cells and the endothelial cells of the choriocapillaris ( Fig. 1C ), plus the extracellular matrix that fi lls the space between these two membranes [reviewed in ( 2, 3 )].The relatively modest compendium of cell types mentioned above, however, belies the amazing, higher order complexity of this sensory tissue. It has been estimated that there are actually about 55 anatomically distinct neuronal cell types in the adult mammalian retina, of which about 22 have been thoroughly characterized with regard to their fi ne structure and functional repertoire [reviewed in ( 4 )]. For example, photoreceptors consist of both rods and cones, with cones being subdivided into two (three in Abstract The vertebrate retina has multiple demands for utilization of cholesterol and must meet those demands either by synthesizing its own supply of cholesterol or by importing cholesterol from extraretinal sources, or both. Unlike the blood-brain barrier, the blood-retina barrier allows uptake of cholesterol from the circulation via a lipoprotein-based/receptor-mediated mechanism. Under normal conditions, cholesterol homeostasis is tightly regulated; also, cholesterol exists in the neural retina overwhelmingly in unesterifi ed form, and sterol intermediates are present in minimal to negligible quantities. However, under certain pathological conditions, either due to an inborn error in cholesterol biosynthesis or as a consequence of exposure to selective inhibitors of enzymes in the cholesterol pathway, the ratio of sterol intermediates to cholesterol in the retina can rise dramatically and persist, in some cases resulting in progressive degeneration that signifi cantly compromises the structure and function of the retina. Background/Aims: The anti-inflammatory activity of 3-aminobenzamide (3-AB) has been shown via histopathology and immunohistochemistry in various colitis models. We aimed to study the effects of 3-AB on tissue mechanical endurance and, associatively, preventing perforation in colitis. Materials and Methods: Thirty male Wistar albino rats were randomly divided into three groups. Rectal saline was administered to Group 1 (sham + saline). Rectal trinitrobenzensulphonic acid was applied to induce colitis in Group 2 (colitis + saline) and Group 3 (colitis + 3-AB). Groups 1 and 2 were treated intraperitoneally with saline (1 ml every 12 hours) and Group 3 was treated with 3-AB (10 mg/kg every 12 hours). After seven days, rats were sacrificed and colon lipid peroxidation levels, the serum tumor necrosis factor alpha (TNF-α) level, bowel bursting pressures, and bowel wall tensions were measured. Results: Bowel bursting pressure in Group 2 was significantly lower than in Groups 1 and 3 (p<0.001 for both groups). Bowel wall tension in Group 2 was significantly lower than in Groups 1 and 3 (p<0.001 for both groups). There were no significant differences between groups for serum TNF-α levels. For lipid peroxidation, malondialdehyde (MDA) levels were increased in Groups 2 and 3 compared to Group 1. Conclusion: 3-AB may aid prevention of perforations that develop in inflammatory bowel disease, requiring surgical treatment.

Hintergrund: Mehrfach ungesÄttigte FettsÄuren (Polyunsaturated Fatty Acids = PUFA) sind ein wichtiger Bestandteil praktisch aller Lipide und damit Bestandteile des zellulÄren Fettdepots sowie Bausteine der Zellmembranen. Zielsetzung: Erstellung eines Positionspapiers fÜr PUFA. Methoden: Systematische Analyse und Bewertung von Humanstudien (prospektive Doppelblindstudien, epidemiologische und retrospektive Studien, kurzfristige biochemische/hÄmatologische Studien («Surrogate Markers»)) der letzten 10 Jahre aus den gÄngigen elektronischen Datenbanken sowie der Angaben von Standardwerken und publizierten Monographien. Ergebnisse und Schlussfolgerungen: Zwei mehrfach ungesÄttigte FettsÄuren, die LinolsÄure und die Alpha-LinolensÄure sind fÜr den Menschen essenziell. Die diversen PUFA sind in verschiedenen Ölen enthalten, vor allem in FischÖl. n-3-PUFA aus FischÖl mÜssen mit Antioxidantien kombiniert werden, damit sie stabilisiert werden und ihre Wirkung entfalten kÖnnen. Die Wahrscheinlichkeit einer unzureichenden Versorgung mit PUFA ist am grÖssten bei Menschen, deren Bedarf erhÖht ist, zum Beispiel bei Infektionen, chronischen Krankheiten oder in Wachstumsphasen. Im Sinne einer prÄventiv wirksamen Zusammensetzung der mehrfach ungesÄttigten FettsÄuren in der Nahrung sprechen sich deutsche, Österreichische und schweizerische ErnÄhrungsgesellschaften dafÜr aus, dass das VerhÄltnis von Alpha-LinolensÄure (n-3) zu LinolsÄure (n-6) auf mindestens 1:5 erhÖht wird. PUFA haben auf verschiedene Organsysteme und bei verschiedenen Erkrankungen eine positive Wirkung: Hautkrankheiten, koronare Herzkrankheit, obstruktive Lungenleiden und rezidivierende Infekte, senkt die postoperative infektiÖse MorbiditÄt. Alpha-LinolensÄure bietet praktische Vorteile gegenÜber FischÖlprÄparaten: Kein unangenehmes Aufstossen fÜr den Patienten und geringerer Bedarf an Stabilisatoren (Antioxidantien) in der galenischen Darreichungsform.
