Nutlin-Loaded Magnetic Solid Lipid Nanoparticles for Targeted Glioblastoma Treatment

Abstract: Aim Glioblastoma multiforme is one of the deadliest forms of cancer, and current treatments are limited to palliative cares. The present study proposes a nanotechnology-based solution able to improve both drug efficacy and its delivery efficiency. Materials & methods Nutlin-3a and superparamagnetic nanoparticles were encapsulated in solid lipid nanoparticles, and the obtained nanovectors (nutlin-loaded magnetic solid lipid nanoparticle [Nut-Mag-SLNs]) were characterized… Show more

“…For example, superparamagnetic iron oxide nanoparticles (SPIONs) interact with external alternated magnetic fields and, therefore, they can be used both for hyperthermia and as contrast agents in magnetic resonance imaging (MRI) . Moreover, the application of an external magnetic field can be exploited to favor the crossing of the BBB of SPIONs, as recently demonstrated . Iron oxide nanoparticles functionalized with catalase were shown to be able to convert ROS into O 2 mitigating hypoxia effects in tumor microenvironment.…”

“…To assess the ability to guide the CM-NCubes across the BBB and towards the glioblastoma site using a static magnetic field, an in vitro fluidic model of the BBB [14] was used. The fluidic model consisted of two parallel channels (top and bottom channel) that were separated by a porous membrane, derived from 6 well falcon transwell inserts (3 µm pore size).…”

“…We also studied the aforementioned magnetic targeting problem using the numerical model that was introduced in literature [ 53 ]. The considered flow channel had a rectangular cross-section (3.8 mm width, 0.4 mm height) and the flow was uniform (thus considered to be fully developed) over a 17 mm span.…”
