Nupr1/Chop signal axis is involved in mitochondrion-related endothelial cell apoptosis induced by methamphetamine

Abstract: Methamphetamine (METH) abuse has been a serious global public health problem for decades. Previous studies have shown that METH causes detrimental effects on the nervous and cardiovascular systems. METH-induced cardiovascular toxicity has been, in part, attributed to its destructive effect on vascular endothelial cells. However, the underlying mechanism of METH-caused endothelium disruption has not been investigated systematically. In this study, we identified a novel pathway involved in endothelial cell apopt… Show more

“…The diabetes patient population is expected to grow in the next decade that will cause huge disease burden to our society. 21,22 In any case, our results provide clear evidence that Nupr1 is involved in DMED. 5,6 In this study, we used a well-established rat model of DMED to examine the molecular mechanisms that contribute to this serious condition.…”

“…19,20 More specifically, Nupr1/C-EBP homologous protein (Chop) pathway participates in the triggering of apoptosis of vascular endothelial cells. 21 Moreover, Nupr1/Chop is characterized as an endoplasmic reticulum stress pathway that mediates apoptosis. 22 However, it remains unknown whether miR-874-3p targets Nupr1 and if this putative interaction could influence DMED.…”

“…Methamphetamine (METH) is one of the most widely abused drugs and the most commonly synthesized illegal drug worldwide (Kiyatkin and Sharma, 2009 ; Cai et al, 2016 ; Li et al, 2017 ). A major target of METH is the central nervous system (CNS), especially the central dopaminergic circuitries (Carmena et al, 2015 ; Huang et al, 2015 ; Chen et al, 2016 ; Mendieta et al, 2016 ).…”

“…It is demonstrated that ER stress plays a key role in the onset and progression of neurodegenerative diseases ( Mercado et al, 2016 ; Thummayot et al, 2016 ). ER stress was also involved in METH-induced umbilical vein endothelial cell injury ( Cai et al, 2016 ). Our previous studies have found that ER stress is related to restraint stress-induced rat kidney injury and hippocampal damage ( Geng et al, 2013 ; Zhang et al, 2014 ).…”
