Number of Dental Stem Cells on Root Canal Dentin after Application of Triple Antibiotic Paste or Calcium Hydroxide: An In Vitro Study

Abstract: Objective The aim of this study was to evaluate the viability of stem cells from exfoliated and deciduous teeth (SHED) on dentin surface treated with triple antibiotic paste or calcium hydroxide. Materials and Methods Nine single-rooted extracted premolars were prepared appropriately and divided into three groups. In group A, the root canals were left empty, a triple antibiotic paste was placed in the root canals of group B, and calcium hydroxide was placed in the root canals of group C. After 1 week… Show more

“…Certain conditions are required to achieve this goal: the presence of stem cells, scaffold and growth factors [11]. The vitality, proliferation and differentiation of stem cells from apical papilla (SCAP) have been shown to be easily affected by inflammation, irrigants and medications in culture [12,13]. The use of devitalizing paste in presented case might have caused harmful adverse effects on these cells.…”

“…Although GFs have short half-life, the binding of bioactive GFs to dentin extracellular matrix proteins protects them from biodegradation and prolongs their life-time span, which could be released in active forms during carious process or following the use of certain dental materials having the ability to cause dentin demineralization; e.g. : Ethylene diamine tetraacetic acid (EDTA), etching adhesives, and calcium hydroxide (5)(6)(7) .…”

“…5 The observed variation in clinical outcomes created a critical gap regarding the effects of these medicaments on stem cell survival and attachment to the root canal of the irrigated dentine. 6 , 7 Some lines of evidence support the use of Ca(OH) 2 , 8 , 9 , 10 , 11 , although a few studies have also indicated that TAP enhanced the attachment of dental pulp stem cells (DPSCs) to the dentine. 12 These authors also found that double antibiotic paste (DAP) and Ca(OH) 2 reduced the proliferation of DPSCs.…”

“… 12 However, these findings were controversial due to the different experimental protocols used and the evaluation of stem cell survival after the short-term application of intracanal medicaments over 3- and 7-day periods. 11 , 12 , 13 …”
