Nucleotide sequence of the region between the 18S rRNA sequence and the 28S rRNA sequence of rat ribosomal DNA

Abstract: The DNA sequence of the intragenic region of the rat 45S ribosomal RNA precursor was determined. This sequence contains 2282 nucleotides and extends from the conserved EcoR I site near the 3' terminus of 18S rRNA to 69 nucleotides downstream of the 5' terminus of 28S rRNA. The sequences corresponding to 18S and 5.8S rRNA were identified by comparison with previously published data. The 5' terminus of rat 28S rRNA was identified by S1 nuclease protection and reverse transcriptase elongation assays. The internal… Show more

“…2). These were noted in a tobacco chloroplast S19 gene (25) and a rat rRNA spacer sequence (24). Whether these sequences participate in mechanisms that regulate the ribosomal gene family and how they might be involved in coordinating the expression of ribosomal protein and rRNA genes is not clear.…”

“…In case RNase treatment was performed, cells were incubated with 100 µg/ml RNase A for 1 hour at 37°C and subsequently washed three times with 2× SSC. Digoxigenin-labeled probes to rRNAs, generated by nick translation from plasmids containing parts of the mature regions of 18S and 28S rRNA, respectively Subrahmanyam et al, 1982), were a kind gift of C. Carvalho and M. Carmo-Fonseca (University of Lisbon, Portugal) (Carvalho et al, 2001). Prior to hybridization, cells were washed with 50% deionized formamide, 2× SSC.…”

“…The 5' end of the 26S rRNA could not be satisfactorily determined. However, the 5' terminal sequences of 26/28S rRNA have been reported for Xenopus, yeast, rat, and mouse (5,6,(20)(21)(22) and in each case the terminal nucleotide is either 2 or 3 bases 5' to a conserved GACCUC sequence. This sequence, which has been implicated in binding with the 3' end of the 5.8S rRNA (20) is also found in the sea urchin rDNA unit at positions 1100-1105.…”

“…The complete sequence of the 26S rDNA has been determined for two species of Saccharomyces (20,21) and for Physarum (28); the first 69 nucleotides of rat 28S rDNA (6), the first 118 nucleotides of Xenopus 28S rDNA (5), and the first 585 nucleotides of mouse 28S rDNA (22) have been published. Comparison of these sequences to the first 505 nucleotides of Lytechinus reveals two regions of high homology.…”
