Nucleotide sequence of the gene encoding novel delta-endotoxin from Bacillus thuringiensis serovar japonensis strain Buibui specific to scarabaeid beetles

Abstract: A new isolate of Bacillus thuringiensis serovar japonensis strain Buibui, which was specific to scarab beetles (M. Ohba et al., Lett. Appl. Microbiol. 14:54, 1992), was shown to have a 130-kDa insecticidal crystal protein (ICP) (H. Hori et al., J. Appl. Bacteriol. 76:307, 1994). ClaI restriction enzyme fragments of total cell DNA of the isolate were cloned into E. coli (Sato et al., Curr. Microbiol. 28:15, 1994). Whole 3480-bp nucleotide sequence of the gene encoding 130-kDa ICP was determined, and the molecul… Show more

“…The sizes of both gene products are within the range calculated for other Cry8 proteins [e.g. Amadio et al, 2013;Ogiwara et al, 1995;Shu et al, 2009]. Alignment of the amino acid sequences of Cry8Kb3 and Cry8Pa3 shows that their sequences share an overall 63% sequence identity (online suppl.…”

“…The Cry8‐type proteins are mainly insecticidal to the larvae of scarab beetles (Sato et al ., 1994; Yu et al ., 2006; Yamaguchi et al ., 2008; Shu et al ., 2009a, b), and some of these proteins also have toxicity against adult beetles (Yamaguchi et al ., 2008). Cry8Ea, a variety of crystal protein, is toxic to Anomala corpulenta larvae, which are important pests in agriculture, horticulture, and forestry (Sato et al ., 1994; Ogiwara et al ., 1995; Huang et al ., 2007; Shu et al ., 2009a). In the present study, both forms of the Cry8Ea1 toxin, i.e.…”

“…It has been well established that (1) the strain HD-1 produces four d-endotoxin proteins (Cry1Aa, Cry1Ab, Cry2 A and Cry2B) (Höfte and Whiteley 1989); (2) the strain T84 A1 has a single protein, Cry1Aa (Ogo et al 1990) and (3) the protein of the strain Buibui belongs to the class Cry8C (Ogiwara et al 1995). Thus, our immunological findings strongly suggest that the proteins of the four strains are not the members of the classes Cry1 A, Cry2 and Cry8.…”
