Nucleotide sequence and organization of potato leafroll virus genomic RNA

Abstract: The nucleotide sequence of the genomic RNA of potato leafroll virus was determined and its genetic organization deduced. The RNA is 5882 nucleotides long and contains 6 open reading frames (ORFs) encoding proteins of 70, 70, 56, 28, 23 and 17 kDa. The putative genes for the coat protein (23 kDa) and the RNA-dependent RNA polymerase (70 kDa) were identified by interviral amino acid sequence homologies. For expression of the different ORFs, translational frameshift and readthrough events are proposed.cDNA clonin… Show more

“…The genomic R N A of PLRV contains a direct repeat of the sequence of the first eight (van der Wilk et al, 1989) or nine nucleotides in the sequence from nucleotides 3377 or 3376 respectively (Fig. 4).…”

“…Analysis of the nucleotide sequence of the genome of PLRV has identified six ORFs (Mayo et al, 1989;van der Wilk et al, 1989;Keese et al, 1990) which are thought to be expressed by a variety of strategies, including initiation at downstream AUG codons, shifting between reading frames during translation, suppression of termination codons and translation of subgenomic RNA containing downstream ORFs (Mayo et al, 1989;Bahner et al, 1990;Keese et al, 1990).…”

“…A second dissimilarity is the absence from PEMV RNA 1 of the highly conserved 17K to 19K protein-encoding reading frame nested within the coat protein gene of luteoviruses. This protein has been postulated to represent the Y-terminal VpG common to both virus groups (Miller et al, 1988a;van der Wilk et al, 1989), and a recent report by Tacke et aI. (1991) has speculated that it may have a role in virus replication, based on its affinity for single-stranded nucleic acids.…”

“…The genomic singlestranded, non-polyadenylated RNA is approximately 6 kb in size and possesses a genome-linked small protein (VPg; Mayo et al, 1982;Harrison, 1984). Sequence analysis has revealed the presence of six large open reading frames (ORFs) in two gene clusters separated by a small intergenic region (Mayo et al, 1989;van der Wilk et al, 1989;Keese et al, 1990;Fig. la).…”
