Nucleolar DNA: the host and the guests

Abstract: Nucleoli are formed on the basis of ribosomal genes coding for RNAs of ribosomal particles, but also include a great variety of other DNA regions. In this article, we discuss the characteristics of ribosomal DNA: the structure of the rDNA locus, complex organization and functions of the intergenic spacer, multiplicity of gene copies in one cell, selective silencing of genes and whole gene clusters, relation to components of nucleolar ultrastructure, specific problems associated with replication. We also review… Show more

“…Its primary function is in the making of ribosomes, and it forms in response to the transcription of the ribosomal DNA (rDNA) and to the recruitment of ribosome biogenesis factors 13 . The pre-ribosomal RNA (pre-rRNA) is transcribed by RNA polymerase I (RNAPI) from the rDNA that is arranged in tandem arrays on chromosomes 13, 14, 15, 21, and 22 in human diploid cells 46 . A series of endo- and exo- nucleolytic cleavages removes the transcribed spacer sequences from the pre-rRNA to release the mature 18S, 5.8S, and 28S rRNAs 7, 8 .…”

“…Light/electronic microscopy, however, reveals that the nucleolus contains three mainly morphologically distinct regions, the fibrillar center (FC), the dense fibrillar component (DFC) and the granular component (GC) [1]. In mammalian cells, several hundred ribosomal DNA genes (rDNA repeats) separated by long intergenic spacers (IGSs) are localized in the FC and are responsible for 47S pre-rRNA transcription [3]. 47S pre-rRNA transcription is dependent on RNA polymerase I (Pol I).…”

“…Structural and functional aspects of various nuclear components and nuclear bodies were reviewed in the March 2016 special issue of HCB "Focus on: the cell nucleus," among them the nucleolus (Smirnov et al 2016;Weipoltshammer and Schöfer 2016). The nucleolus is the largest nuclear body, and its main function is in ribosome biogenesis.…”
