“…The clock influences several important hallmarks and pathways in GBM. This implies that combining current emerging targeted therapies such as therapies targeting energy metabolism, angiogenesis, the PI3K/AKT/mTORC pathway 2 or immune therapies 63 , 64 with chronotherapy could improve their performance. Future research should therefore focus on exploring feasible methodologies to characterize the relationship between those GBM features and the clock in individual patients, and how to exploit it therapeutically.…”

“…Also, two studies found evidence for exosomes in mediating the interplay of the clock and tumour promoting inflammation, which makes them another interesting target 28 , 32 . As reviewed by Pinheiro et al 63 and Dain et al 64 there are different possibilities to reprogram immune cells such as cytokine therapy and immune checkpoint inhibitors. Approaches targeting IFNγ (Interferon gamma), TNFα, or the PD-1 (Programmed Cell Death 1)/PDL-1 (Programmed Cell Death Ligand 1) axis may provide excellent opportunities for combination with clock-based strategies, as those targets are controlled by the clock 63 .…”

“…One of the most heavily researched brain cancers and most common is glioblastoma multiforme, an aggressive type of brain cancer that is formed from astrocytes ( Ray-Chaudhury and Ray, 2010 ). Treatment of these cancers is challenging because the tumor is very malignant and it is most common in older adults, who may not be able to tolerate aggressive chemotherapy ( Dain and Zhu, 2023 ). The current treatment plan for someone with glioblastoma cancer includes chemotherapy, radiation therapy, surgery to excise the tumor, experimental clinical trials, tumor treating fields (TTF) therapy, or palliative care, however survival rates are still very low ( mayoclinic, 2022a ).…”

“…There are no known risk factors for GBM, and it occurs without warning signs. Furthermore, its incidence increases with age, with white ethnicity being more commonly affected than black ethnicity, and males being more affected than females ( Newton et al, 2018 ; Grochans et al, 2022 ; Dain and Zhu, 2023 ). Considering the invasive nature of GBM and resistance to therapies, recurrence is observed after treatments ( Sareen et al, 2022 ).…”
