Nucleic Acid Carriers Based on Precise Polymer Conjugates

Abstract: Polymer polydispersity, random conjugation of functional groups, and poorly understood structure–activity relationships have constantly hampered progress in the development of nucleic acid carriers. This review focuses on the synthetic concepts for the generation of precise polymers, site-specific conjugation strategies, and multifunctional conjugates for nucleic acid transport. Dendrimers, defined peptide carriers, sequence-defined polyamidoamines assembled by solid-phase supported synthesis, and precise lipo… Show more

“…7,11,15,16,22,36 Receptor-mediated endocytosis can enhance the cellular uptake of nanoparticles. 11,16,22,31 In our experiments, the U-87 cells could not be transfected effectively with unmodified PEI nanoparticles at an N/P ratio of 10 ( Figures 3 and 4). In general, the VTW peptide facilitated the transfection of PEI nanoparticles into U-87 cells via receptor-mediated uptake, while it reduced the cytotoxicity (Figure 4) due to less residual positive charge on the surface of the nanoparticles.…”

“…Many techniques, including purification, preparation, and chemical modification, have been used to improve the properties of nanoparticles. 16,31 A heterobifunctional crosslinker, SPDP, has an amine-reactive radical called the N-hydroxysuccinimide ester and a sulfhydryl-reactive radical called 2-pyridyldithio. [32][33][34] This crosslinker guaranteed that PEI-PDT and VTW were conjugated accurately.…”

“…Currently, the use of polymer for siRNA or drug delivery have been reported extensively (Nimesh et al, 2011;Singha et al, 2011;Troiber and Wagner, 2011;Duncan, 2011). RNA is a polymer (polynucleic acid).…”

“…Various strategies for RNA delivery have been explored, such as the application of lipid-based delivery systems (lipoplexes and liposomes) [4][5][6] , nanoparticles [7][8][9] , viruses 10 , peptide-based systems 7 , polymers [11][12][13][14] and gold-based materials [15][16][17][18] .…”
