Nuclear Translocation of a Clusterin Isoform Is Associated with Induction of Anoikis in SV40‐Immortalized Human Prostate Epithelial Cells

Abstract: Clusterin gene expression is potently induced in experimental models in which apoptosis is activated, such as rat prostate involution following castration. Nevertheless, its precise physiological role has not yet been established, and both anti-apoptotic and pro-apoptotic functions have been suggested for this gene. Clusterin expression level depends on cell proliferation state, and we recently showed that its over-expression inhibited cell cycle progression of SV40-immortalized human prostate epithelial cells… Show more

“…Under these condition, as previously reported, 35 KSFM caused a marked inhibition of cell growth rate when compared to standard culture conditions, inducing anoikis (cell detachment followed by chromatin condensation). Addition of either 1.8 mM Ca 2 þ or the caspase inhibitor benzyloxycarbonyl-Val-Ala-Asp-flu-oromethylketone (z-VAD-fmk, 1 mM) to KSFM medium completely rescued PNT1A cells and prevented cell detachment and anoikis induction ( Figure 1a).…”

“…The consequent change in the pCLU/ sCLU ratio might take part in the processes leading GTE to counteract PCa progression (19,(26)(27)(28). The above hypothesis is currently under investigation in our laboratory.…”

“…CLU is considered an enigmatic protein because of the difficulties in the definition of its precise functions; at the moment, an important debate concerns its implication in carcinogenesis, particularly in prostate cancer development. On this issue, the data currently available are controversial because CLU has been suggested as being either a proapoptotic or a prosurvival factor (3)(4)(5)(6)(7)(8)(9).…”
