Nuclear genes involved in mitochondrial diseases caused by instability of mitochondrial DNA

Abstract: Mitochondrial diseases are defined by a respiratory chain dysfunction and in most of the cases manifest as multisystem disorders with predominant expression in muscles and nerves and may be caused by mutations in mitochondrial (mtDNA) or nuclear (nDNA) genomes. Most of the proteins involved in respiratory chain function are nuclear encoded, although 13 subunits of respiratory chain complexes (together with 2 rRNAs and 22 tRNAs necessary for their translation) encoded by mtDNA are essential for cell function. n… Show more

“…As mtCN of individuals is known to be correlated with mitochondrial functions regulated by the nDNA genes 3,4 , we also investigated the mtCN-nDNA genotype association. Each mtCN of the individual was estimated by the WGS depth.…”

“…Hundreds of mitochondria reside in a single cell and each mitochondrion has several mtDNA molecules in itself. Furthermore, 1000-2000 mitochondria-related genes in nuclear DNA (nDNA) have been identified, which regulate gene expressions and mtDNA copy numbers (mtCNs) via interactions with mtDNA [3][4][5][6] .…”

“…A priori, we expect that mitochondrially encoded gene expression would be positively correlated with mtDNA-CN. Likewise, multiple nuclear encoded genes are involved in the regulation of mtDNA replication, and thus, expression levels of these genes are expected to be correlated with mtDNA-CN [23,24]. We therefore evaluated the associations between mtDNA-CN and expression of these two classes of genes, correcting for cohort, sample ischemic time, genotyping PCs, age, race, and surrogate variables derived from RNA-sequencing data to capture known and hidden confounders (Supp.…”
