Nuclear Functions of Nucleolin through Global Proteomics and Interactomic Approaches

Abstract: Nucleolin (NCL) is a major component of the cell nucleolus, which has the ability to rapidly shuttle to several other cells' compartments. NCL plays important roles in a variety of essential functions, among which are ribosome biogenesis, gene expression, and cell growth. However, the precise mechanisms underlying NCL functions are still unclear. Our study aimed to provide new information on NCL functions via the identification of its nuclear interacting partners. Using an interactomics approach, we identified… Show more

“…To gain more insight into the potential implication of Ncl in spliceosome assembly/function, in this work, we studied the subcellular co-localization and interaction of Ac-Ncl with a component of catalytically activated/active spliceosome, P-SF3b155 (phosphorylated SF3B1). Together with the re-analysis of the splicing components found in our previous proteomic data [23], our data suggest that Ncl interact mostly with the pre-catalytic B complex and could probably participate in the assembly of the pre-catalytic spliceosomes.…”

“…Gene ontology of the purified proteins identify also 45 proteins implicated in RNA metabolism (ribosomal RNA excluded) including RNA stability, RNA nuclear export, pre-mRNA splicing. Interestingly, most of these proteins were still associated with Ncl after RNAse treatment, suggesting a direct protein-protein association [23].…”

“…Nucleolin (NCL) is a highly conserved, multifunctional RNA binding protein (RBP) implicated in several cellular processes such as chromatin remodeling [1], rRNA synthesis [2], mRNA processing [3], ribosome assembly, and nucleo-cytoplasmic transport [4]. Elevated levels of NCL are present in highly proliferative cells including a variety of tumors [5][6][7][8][9].…”
