Nuclear Factor-1-X Regulates Astrocyte-specific Expression of the α1-Antichymotrypsin and Glial Fibrillary Acidic Protein Genes

Abstract: Discrete tissue-specific changes in chromatin structure of the distal serpin subcluster on human chromosome 14q32.1 allow a single gene encoding ␣ 1 -antichymotrypsin (ACT) to be expressed in astrocytes and glioma cells. This astrocyte-specific regulation involves activatory protein-1 (AP-1) because overexpression of dominantnegative c-jun(TAM67) abolishes ACT expression in glioma cells. Here we identify a new regulatory element, located within the ؊13-kb enhancer of the ACT gene, that binds nuclear factor-1 (… Show more

“…For example, NFIs have been demonstrated to bind to the promoters of neuronal-and glialspecific genes and to regulate their activity in vitro (Copertino et al, 1997;Behrens et al, 2000;Bisgrove et al, 2000;Wang et al, 2004;Cebolla and Vallejo, 2006;Gopalan et al, 2006), implying direct gene regulation by the NFI transcription factors. Moreover, our data clearly indicate that, in addition to regulating astrocytic differentiation within the telencephalon, NFIA also acts to negatively regulate the Notch signaling pathway, in part via direct repression of Hes1.…”

“…1E,F). NFIX has previously been linked to the regulation of astrocyte-specific genes including Gfap, brain fatty acid-binding protein, and α1-antichymotrypsin (Gopalan et al 2006;; however, our findings indicate that NFIX may also regulate transcriptional activity within neural progenitor cells in the developing hippocampus.…”

“…Nfix −/− mice also display severe neurological phenotypes , and NFIX has previously been implicated in driving the expression of astrocytic genes during neural development (Gopalan et al 2006;. However, the mechanism by which NFIX regulates morphogenesis of the nervous system in vivo remains undefined.…”
