Nuclear Export and Import of Human Hepatitis B Virus Capsid Protein and Particles

Abstract: It remains unclear what determines the subcellular localization of hepatitis B virus (HBV) core protein (HBc) and particles. To address this fundamental issue, we have identified four distinct HBc localization signals in the arginine rich domain (ARD) of HBc, using immunofluorescence confocal microscopy and fractionation/Western blot analysis. ARD consists of four tight clustering arginine-rich subdomains. ARD-I and ARD-III are associated with two co-dependent nuclear localization signals (NLS), while ARD-II a… Show more

“…Heterokaryon assays were performed as described previously (19). Briefly, at 24 h posttransfection, human Huh7 cells were treated with trypsin, and 10 6 Huh7 cells were mixed with 2 Ï« 10 6 untransfected mouse NIH 3T3 cells, before being seeded on glass coverslips in a 6-cm dish.…”

“…For assembled capsids at least, nuclear import requires phosphorylation (Kann et al, 1999). While the nuclear transport capacity of core is well established, a recent publication also reported nuclear export of core protein, which was thought to be implicated in export of HBV specific transcripts by the TAP/NxF1 pathway (Li et al, 2010).…”
