Nuclear DNA Content as an Index Character Discriminating Taxa in the Genus Petunia sensu Jussieu (Solanaceae)

Abstract: Nuclear DNA contents over the total range of the genus Petunia sensu Jussieu comprising 20 taxa of Petunia sensu Wijsman (2n 2x 14) and 32 taxa of Calibrachoa (2n 2x 18) were estimated by¯ow cytometry after staining the nuclei with propidium iodide (PI) or 4H ,6-diamidino-2-phenylindole (DAPI). With respect to nuclear DNA content, taxa of Petunia sensu Wijsman seemed to be homogeneous (2C 2 . 60 to 3 . 41 pg), but Calibrachoa taxa were clearly separated into two groups: (1) C. parvi¯ora and C. pygmaea (1 . 56 … Show more

“…After filtration through a 30-μm nylon mesh, crude nuclear samples were stained with 0.8 ml DAPI solution containing 10 mM Tris, 50 mM sodium citrate, 2 mM MgCl 2 , 1% (w/v) PVP K-30, 0.1% (v/v) Triton X-100, and 2 mg l -1 DAPI (pH 7.5) (Mishiba et al 2000 Relative DNA content was estimated by the ratio between the fluorescence intensity of the sample and internal standard: sample relative DNA content = sample peak mean / standard peak mean. The relative DNA content was then calculated for each sample as the average of the three replications.…”

“…After filtration through a 30 μm nylon mesh, crude nuclear samples were stained with 0.7 mL of DAPI (4',6-diamidino-2-phenylindole) solution containing 10 mM Tris, 50 mM sodium citrate, 2 mM MgCl 2 , 1% (w/v) polyvinylpyrrolidone (PVP) K-30, 0.1% (v/v) Triton X-100, and 2 mg·L −1 DAPI (pH 7.5) (Mishiba et al, 2000). After incubation for 30 s at room temperature, nuclear fluorescence was measured using a ploidy analyzer (Partec PA; Partec).…”

“…To induce a larger-flowered phenotype of petunia, it is desirable to identify G and elucidate the underlying molecular mechanism of flower enlargement and of these pleiotropic effects. For this purpose, a genetic approach using a linkage map (Bossolini et al, 2011) might be effective, although large genome size (1200-1500 Mbp, Mishiba et al, 2000) may be a barrier to the identification of G. It may be useful to perform transcriptome analysis between gg and Gg plants based on ample EST data deposited in public databases such as the DNA Data Bank of Japan (DDBJ, http://www.ddbj.nig.ac.jp/) to specify gene regulation networks influenced by G.…”
