“…Fax: +81-52-789-4087; E-mail: koba@agr.nagoya-u.ac.jp progeny of the cross between ABPU1 (biA1 pyrG89; wA3; argB2; pyroA4) and FGSC A57 (yA2; pyroA4; amyR1/malA1), 27) and SA1 was made by a cross between ÁamyR103 and DAT1 in this study. The A. nidulans strains were grown at 37 C in standard minimal medium 31) supplemented with appropriate requirements unless stated otherwise. Transformation of A. nidulans was performed by methods described elsewhere 32) except that protoplasts were generated using a mixture of Yatalase (Takara, Kyoto, Japan) and Lysing enzyme (Sigma, Tokyo, Japan) at final concentrations of 3 mg/ml and 0.3 mg/ml respectively.…”

“…Strains were grown at 37 C in YGuu medium [0.5% yeast extract (Difco), 1% glucose, 0.1% trace element, 10 mM uridine, and 10 mM uracil] or minimal medium (MM). 19) MM was supplemented with biotin at 0.02 mg/ml, pyridoxine at 0.5 mg/ml, and 10 mM uridine and 10 mM uracil when necessary, and each supplement added was indicated by a single lowercase letter after the medium name. To test the effect of osmolarity, 1/2 YGuu (0.25% yeast extract, 0.5% glucose, 0.1% trace element, 10 mM uridine, and 10 mM uracil) and YGuu plus 1.0 M sorbitol were used.…”

“…nidulans is a homothallic organism but, at the origin of each cleistothecium, each of the two nuclei can behave as either the male or the female gamete. This is shown by the behaviour of both mitochondrial and maternal-effect nuclear genes (Apirion, 1963;Roberts and Orias, 1973;Rowlands and Turner, 1973;Gunatilleke et al, 1975). In particular, when two strains carrying two different alleles of a mitochondrial gene are crossed, all ascospores in all asci in a given cleistothecium carry exclusively, without exception, one and the same allele.…”
