NUAK2 silencing inhibits the proliferation, migration and epithelial‑to‑mesenchymal transition of cervical cancer cells via upregulating CYFIP2

Abstract: NUAK family kinase 2 (NUAK2) has been reported to be involved in various cancer cell processes, including proliferation, apoptosis and invasion, by targeting multiple genes. However, to the best of our knowledge, its biological function in cervical cancer (CC) has not yet been elucidated. Therefore, the present study aimed to measure the expression of NUAK2 and to evaluate its functions in CC. The expression levels of NUAK2 and cytoplasmic FMRP-interacting protein 2 (CYFIP2) were detected in CC tissues and cel… Show more

“…We also found a tumor suppressor OPCML, which is already recognized in epithelial ovarian cancer (71,85). Moreover, a new potential inhibitor, CYFIP2, which promotes cell growth, cell migration (72), and chemoresistance in pancreatic neuroendocrine neoplasms cancer (77) by activation of the Akt signal (73), showed potential for therapeutic applications among other malignancies (74,75) or metastasis cases (76).…”

“…Western blotting was performed according to a previous study (Li et al., 2021). The tissues or cells were incubated with RIPA buffer (Beyotime) and then centrifuged at 12,000 g for 20 min.…”

“…2.5 | Cell counting kit-8 (CCK-8) assay CCK-8 assay was conducted according to a previous study (Li et al, 2021). HLECs were placed into the 96-well plate and reacted with 10 μL CCK-8 solution (Beyotime) for 2 h. Lastly, the absorbance was measured and recorded at 450 nm by a microplate reader.…”

“…Protein expression in lysate or supernatant was analyzed by western blot as previously described (Li et al, 2021). The proteins were extracted using RIPA lysis buffer (Beyotime, Shanghai, China), separated by SDS‐PAGE, and electrophoretically transferred onto PVDF membranes (Millipore, Billerica, MA, USA).…”

“…NUAK2 is a member of the AMPK protein kinase family and is recognized as an indispensable activator for YAP‐driven overgrowth, stem cell expansion, and tumorigenesis (Camargo et al, 2007). In addition, NUAK2 can be inhibited by the tumor suppressor hepatic kinase B1, as well as by NF‐κB, which blocks the activation of death receptor signaling (Li et al, 2021). NUAK2 was shown to be increased in BC, was an independent prognostic indicator of survival for this cancer, and knockdown of NUAK2 could inhibit BC cell proliferation and tumor‐forming ability (Chen et al, 2022).…”
