The papillomavirus E2 protein is involved in the maintenance of persistent infection and known to bind either to cellular factors or directly to mitotic chromosomes in order to partition the viral genome into the daughter cells. However, how the HPV-16 E2 protein acts to facilitate partitioning of the viral genome remains unclear. In this study, we found that serine 243 of HPV-16 E2, located in the hinge region, is crucial for chromosome binding during mitosis. Bromodomain protein 4 (Brd4) has been identified as a cellular binding target through which the E2 protein of bovine papillomavirus type 1 (BPV-1) tethers the viral genome to mitotic chromosomes. Mutation analysis showed that, when the residue serine 243 was substituted by glutamic acid or aspartic acid, whose negative charges mimic the effect of constitutive phosphorylation, the protein still can interact with Brd4 and colocalize with Brd4 in condensed metaphase and anaphase chromosomes. However, substitution by the polar uncharged residues asparagine or glutamine abrogated Brd4 and mitotic chromosome binding. Moreover, following treatment with the inhibitor JQ1 to release Brd4 from the chromosomes, Brd4 and E2 formed punctate foci separate from the chromosomes, further supporting the hypothesis that the association of the HPV-16 E2 protein with the chromosomes is Brd4-dependent. In addition, the S243A E2 protein has a shorter half-life than the wild type, indicating that phosphorylation of the HPV-16 E2 protein at serine 243 also increases its half-life. Thus, phosphorylation of serine 243 in the hinge region of HPV-16 E2 is essential for interaction with Brd4 and required for host chromosome binding.

RESUMOObjetivo: identificar os fatores de risco à infecção pelo Papilomavírus Humano (HPV) associados aos comportamentos e atitudes de adolescentes e jovens de uma unidade escolar de Ensino Médio do Rio de Janeiro. Método: estudo quantitativo, descritivo, realizado de maio a novembro de 2012, com 128 mulheres entre 15 e 24 anos. Os dados foram coletados por questionário e receberam tratamento estatístico descritivo, sendo destacados em variáveis demográficas, econômicas e comportamentais. O estudo foi aprovado pelo protocolo n o 030/2011. Resultados: os dados apontaram haver resistência ao uso de preservativo nas relações sexuais, sendo que 81,3% das mulheres nunca utilizaram camisinha nas relações. O estudo também destacou como uma das características entre as participantes o uso de preservativo somente na primeira relação. O preservativo feminino não foi utilizado pelo grupo estudado. Conclusão: o estudo mostra dados epidemiológicos e questões socioculturais importantes que indicam a necessidade de investigações aprofundadas, além da atuação do enfermeiro em novas estratégias educativas visando à efetividade na mudança de comportamento e atitude desses segmentos populacionais. Palavras-chave: Saúde da mulher; Papilomavírus Humano; fatores de risco; preservativos.ABSTRACT Objective: to identify risk factors for human papillomavirus (HPV) infection associated with behaviors and attitudes of adolescents and young people at an upper secondary school in Rio de Janeiro. Method: in this quantitative, descriptive study conducted from May to November 2012, data were collected by questionnaire from 128 women from 15 to 24 years old, and received descriptive statistical treatment, which highlighted demographic, economic and behavioral variables. The study was approved by the research ethics committee of the Anna Nery School of Nursing, São Francisco de Assis School Hospital, Rio de Janeiro Federal University (protocol No. 030/2011). Results: the data indicated resistance to condom use in sexual relations: 81.3% of the women never used condoms in their relationships. Another of the participants' characteristics highlighted by the study was the use of a condom in the first relation only. Female condoms were not used by the group studied. Conclusion: the study shows epidemiological data and important sociocultural issues that indicate the need for in-depth research, as well as the nurses' playing a role in new educational strategies designed to be effective in changing behavior and attitudes in these population segments.

BackgroundHuman papillomavirus type 16 (HPV 16) E2 protein is a multifunctional DNA-binding protein. HPV 16 E2 regulates many biological responses, including DNA replication, gene expression, and apoptosis. The purpose of this study was to investigate the relationship among the receptor for globular heads of the human C1q (gC1qR) gene expression, HPV 16 E2 transfection and apoptosis regulation in human cervical squamous carcinoma cells (C33a and SiHa).MethodsgC1qR expression was examined in C33a and SiHa cells using real-time PCR and Western blot analysis. Apoptosis of C33a and SiHa cells was assessed by flow cytometry. C33a and SiHa cell viability, migration and proliferation were detected using the water-soluble tetrazolium salt (WST-1) assay, a transwell assay and 3H-thymidine incorporation into DNA (3H-TdR), respectively.ResultsC33a and SiHa cells that were transfected with a vector encoding HPV 16 E2 displayed significantly increased gC1qR gene expression and p38 mitogen-activated protein kinase (p38 MAPK)/ c-jun N-terminal kinase (JNK) activation as well as up-regulation of cellular apoptosis, which was abrogated by the addition of gC1qR small interfering RNA (siRNA). Furthermore, the changes in C33a and SiHa cell viability, migration and proliferation that were observed upon HPV 16 E2 transfection were abrogated by SB203580 (a p38 MAPK inhibitor) or SP600125 (a JNK inhibitor) treatment.ConclusionThese data support a mechanism whereby HPV 16 E2 induces apoptosis by silencing the gC1qR gene or inhibiting p38 MAPK/JNK signalling in cervical squamous cell carcinoma.
