Novel Variants of the IL-10 Receptor 1 Affect Inhibition of Monocyte TNF-α Production

Abstract: IL-10-deficient mice exhibit spontaneous enterocolitis and other symptoms akin to Crohn’s disease, indicating that IL-10 might regulate normal physiology in the gut. However, clinical trials with IL-10 in Crohn’s disease were disappointing, although some patients showed healing of intestinal mucosa. This study searched for genetic polymorphisms within the IL-10 pathway. We decided to screen for mutations of the IL-10R1 cDNA in healthy volunteers and Crohn’s disease patients and identified two novel variants: a… Show more

“…37 Specifically, monocytes from homozygous mutant individuals were not capable to completely downregulate LPSinduced TNF-alpha production upon high doses of IL-10. Also, the ID50 (the IL-10 concentration that caused a 50% reduction of LPS-induced TNF-alpha production) was significantly higher in monocytes from homozygous mutant individuals.…”

“…Exclusion criteria were alcohol abuse (weekly ethanol consumption of more than 50 g for males Genotyping of IL-10R1 variants Two allele-specific polymerase chain reaction (PCRs) were used for detection of IL-10R1 SNP3 and SNP4 in genomic DNA as described. 37 The PCR products of the SNP3 reaction are 464-bp (control PQ-138), 337-bp (wildtype AQ-138) and 183-bp (variant PB-138). The PCR products of the SNP4 reaction are at 515-bp (control PQ-330), 318-bp (wild-type AQ-330) and 248-bp (variant PB-330).…”

“…These variants were not associated with CD but affected IL-10-dependent inhibition of tumor necrosis factor-a production in monocytes. 25 The aim of this study was to investigate the effect of these variants on IL-10 signal transduction, the distribution in various populations and their significance for UC susceptibility.…”
