Novel small molecule modulators of plant growth and development identified by high-content screening with plant pollen

Abstract: BackgroundSmall synthetic molecules provide valuable tools to agricultural biotechnology to circumvent the need for genetic engineering and provide unique benefits to modulate plant growth and development.ResultsWe developed a method to explore molecular mechanisms of plant growth by high-throughput phenotypic screening of haploid populations of pollen cells. These cells rapidly germinate to develop pollen tubes. Compounds acting as growth inhibitors or stimulators of pollen tube growth are identified in a scr… Show more

“…Although the mechanisms by which these small compounds induce reprogramming are unknown, they have shown activity as epigenetic modulators, autophagy modulators and inhibitors of various signalling pathways. The potential of the screening of chemical libraries of small molecules in plant biology research has been reported, although its efficient application in plants is still quite limited (Chuprov-Netochin et al, 2016;Hicks and Raikhel, 2012).…”

“…However, a large number of molecular components governing signal transduction are yet to be discovered and often remain elusive because of lethality and redundancy issues of gene families . To overcome the complexity of whole plant/organ systems with different cell types, single cell models are increasingly used to dissect signalling pathways in combination with quantitative high‐throughput screening systems …”

“…7,8 To overcome the complexity of whole plant/organ systems with different cell types, single cell models are increasingly used to dissect signalling pathways in combination with quantitative high-throughput screening systems. [9][10][11][12] In plants, guard cells are ultimately isolated from the symplast and therefore have been well-established as a single cell model system. [13][14][15] A pair of guard cells forms the stomatal pore evolved to facilitate photosynthetic gas exchange and water transpiration across the leaf epidermal barrier.…”

“…The other nucleophiles such as pyrrole, acetylacetone, malononitrile, pyridine-2-thiol, methimazole and azidotrimethylsilane (entries 12-17, Table 3) could also be easily converted to the target products under the optimized reaction conditions with excellent product yields. Owing to the potential biological activities of compounds bearing xanthen-9-yl groups, [46][47][48] the current efficient catalytic methods for the preparation of variant xanthen-9-yl derivatives would be valuable.…”
