Novel Signal Transduction Pathway for Luteinizing Hormone and Its Interaction with Insulin: Activation of Janus Kinase/Signal Transducer and Activator of Transcription and Phosphoinositol 3-Kinase/Akt Pathways

Abstract: The actions of LH are mediated through a single class of cell surface LH/human chorionic gonadotropin receptor, which is a member of the G protein-coupled receptor family. In the present study we showed that LH induced rapid tyrosine phosphorylation and activation of the Janus kinase 2 (JAK2) in rat ovary. Upon JAK2 activation, tyrosine phosphorylation of signal transducer and activator of transcription-1 (STAT-1), STAT-5b, insulin receptor substrate-1 (IRS-1), and Src homology and collagen homology (Shc) were… Show more

“…FSH stimulated PI3K/AKT signal transduction in many cells, including granular cells, Sertoli cells, and oocytes [39][40][41]. FSH has also been shown to activate ERK and p38 mitogen-activated protein kinase (MAPK) in granulose cells [42,43].…”

“…Furthermore, other hormones that act through GPCRs, including angiotensin II (AII), vasopressin, and luteinizing hormone (LH) can induce tyrosine phosphorylation of cytoplasm proteins (Marrero et al 1995;Saad et al 1995;Velloso et al 1996;Gallo-Payet and Guillon 1998;Carvalho et al 2003). AII and LH activate the cytosolic protein tyrosine kinase Janus kinase 2 (JAK2) and probably use this kinase to induce intracellular protein tyrosine phosphorylation.…”

“…Activation of the JAK/STAT system in some non-ovarian cells in response to leptin, LH and insulin (Carvalho et al 2003) provides indirect evidence that STAT may mediate the effect on some hormones from the ovary. Direct evidence, especially in relation to ovarian cells, was absent.…”

“…Furthermore, activation of STAT1 phosphorylation after addition of leptin to human keratinocytes (Groschl et al 2005), and after LH and insulin treatment of rat ovary (Carvalho et al 2003), indicates that STAT1 can potentially mediate the effects of some hormones on ovarian cells. Nevertheless, there is no direct evidence for an involvement of STAT in mediating hormone action.…”
