Novel role for the double‐stranded RNA‐activated protein kinase PKR: modulation of macrophage infection by the protozoan parasiteLeishmania

Abstract: The evolution of Leishmania infection depends on the balance between microbicidal and suppressor macrophage functions. Double-stranded RNA (dsRNA)-activated protein kinase R (PKR), a classic antiviral protein, is able to regulate a number of signaling pathways and macrophage functions. We investigated the possible role of PKR in the modulation of Leishmania infection. Our data demonstrated that Leishmania amazonensis infection led to PKR activation and increased PKR levels. Consistently, in macrophages from PK… Show more

“…Despite the fact that type I IFNs are mainly known for their antiviral role, increasing evidence attests to the involvement of type I IFNs in bacterial and parasitic infection (24)(25)(26)(27)29). Our findings are consistent with earlier reports suggestive of a disease-exacerbatory role for type I interferons in New World Leishmania species (33)(34)(35).…”

“…During parasite infection, type I IFNs show more variable effects, being either protective or detrimental for the host, depending on the dose, timing of administration, and the parasite species (28,29). For L. major, type I IFNs have been associated with control (30)(31)(32), whereas for other species, especially the New World species L. amazonensis and L. braziliensis, IFN-β has been associated with promotion of parasite survival and/or disease (33)(34)(35).…”

“…PKR is known to have antiviral properties due to its role in translational control. However, a possible role of PKR in protozoan parasitic infection (Leishmania) has been reported recently (41). The observation of an association between Toxoplasma infection and PKR has not been reported previously.…”

“…The region showing the highest excessive European ancestry is located on 3q13, which only harbors LSAMP, a candidate of the tumor suppressor gene in human osteosarcomas, and is associated with coronary artery disease (Kresse et al 2009;Yen et al 2009). Interestingly, EIF2AK2, involved in influenza infection pathways (McAllister et al 2010;Pereira et al 2010), is located ;200 Kb away from the peak (2p22) showing the second highest excessive European ancestry, which suggested the possible difference between African and European resistance to influenza. PACRG, located in 6q26, is associated with Parkinson's disease.…”
