Novel QUEST MRI In Vivo Measurement of Noise-induced Oxidative Stress in the Cochlea

Abstract: Effective personalized therapeutic treatment for hearing loss is currently not available. Cochlear oxidative stress is commonly identified in the pathogenesis of hearing loss based upon findings from excised tissue, thus suggesting a promising druggable etiology. However, the timing and site(s) to target for anti-oxidant treatment in vivo are not clear. Here, we address this long-standing problem with QUEnch-assiSTed Magnetic Resonance Imaging (QUEST MRI), which non-invasively measures excessive production of … Show more

“…Jerk amplitudes are used to elicit far-field potential responses generated by irregular primary vestibular afferent discharges [ 152 , 153 ]. Studies have shown that vestibular damage can occur due to brief exposure to elevated sound levels and sustained exposure to low-frequency continuous sound at more moderate levels [ 154 , 155 , 156 , 157 ]. A study of 258 military personnel found that vestibular damage from intense noise exposure was corelated with asymmetric NIHL [ 138 ].…”

“…Free radicals are paramagnetic, and during times of OS they cause shortening of spin-lattice relaxation time (T1) and thus prolongation of the spin-lattice relaxation rate (R1). By assessing a reduction in R1 after a quench with anti-oxidant treatment during the scan (e.g., administration of alpha-lipoic acid) this weights the specific contribution of ROS to the increased R1 ( 56 ). However, relaxation rates may be impacted by factors that are not related to ROS—such as water and oxygen content ( 57 , 58 ).…”

“…This method can potentially broaden our understanding of the regional specificity of oxidative stress, can help evaluate novel antioxidant treatments in animal models or patients with NDs, and may guide personalized treatment decisions in the future. QUEST-MRI has already been successfully applied in rodent models and humans [170,171]. However, studies on patients with NDs are currently lacking.…”
