Novel polyoxins generated by heterologously expressing polyoxin biosynthetic gene cluster in the sanN inactivated mutant of Streptomyces ansochromogenes

Abstract: BackgroundPolyoxins are potent inhibitors of chitin synthetases in fungi and insects. The gene cluster responsible for biosynthesis of polyoxins has been cloned and sequenced from Streptomyces cacaoi and tens of polyoxin analogs have been identified already.ResultsThe polyoxin biosynthetic gene cluster from Streptomyces cacaoi was heterologously expressed in the sanN inactivated mutant of Streptomyces ansochromogenes as a nikkomycin producer. Besides hybrid antibiotics (polynik A and polyoxin N) and some known… Show more

“…In addition to being outstanding secondary metabolite producers, including of antibacterial and antifungal peptides (Harir et al, 2018), in their own right, Streptomyces species also offer many potential advantages as hosts for the expression and secretion of proteins (Baltz, 2010). In a small number of cases they have also been used for the heterologous production of AFPs (Li et al, 2012;Roldán-Tapia et al, 2017) (Table 4).…”

“…The entire polyoxin and nikkomycin biosynthetic gene clusters were ligated with integrated vector pSET152 to generate pPOL and pNIK, respectively [34,35]. Then they were introduced into M145 and rimP-SCDM to generate recombinant strains M145/pPOL, rimP-SCDM/pPOL, M145/pNIK and rimP-SCDM/pNIK.…”

“…Reports in the literature show that S. ansochromogenes produces a number of antibiotic agents belonging to different chemical classes and with different structures (Chen et al, 2000;Tan et al, 2002;Liu et al, 2005). According to Li et al (2012), S. ansochromogenes has strong antifungal activity that arises from chemical components such as nikkomycin Z and polyoxin, which are nucleoside peptide antibiotics that are strong competitive inhibitors of chitin synthesis.…”
