Novel Oxazolidinone Antibacterial Analogues with a Substituted Ligustrazine C‐ring Unit

Abstract: A series of novel oxazolidinone compounds with a substituted ligustrazine C-ring unit and different substituted groups at the C-5 side chain were designed and synthesized using linezolid as a lead and based on a scaffold hopping strategy. Their antibacterial and anti-inflammatory activities were evaluated. The results of in vitro antibacterial assays showed that all fourteen target compounds displayed potent activity against Gram-positive pathogens, particularly 8b, 13b, 14a, 14b, 15a, and 15b. Moreover, 14a a… Show more

“…Therefore, that study concluded that LZD suppresses the production of TNF-α and IFN-γ without exerting any inhibitory effect on endotoxin production by bacteria (Takahashi et al, 2010). (Chen et al, 2015) compared the effects of a series of novel oxazolidinone compounds and LZD on the production of inflammatory mediators (NO, IL-6, and TNF-α) in LPS-stimulated murine macrophages. They found that some of these oxazolidinone compounds exhibited a more significant inhibitory effect than LZD.…”

“…Recent studies showed that natural products or herbal components provide a new direction for drug research and development . Tetramethylpyrazine (TMP), an effective blood circulation drugs, exhibits an ample array of pharmacological activities such as antithrombotic , antiangina and antibacterial properties . We previously reported that TMP alleviated carbon tetrachloride (CCl 4 )‐induced liver fibrosis in vivo and inhibited platelet‐derived growth factor‐β receptor (PDGF‐βR) pathway and NLRP3 inflammasome expression of hepatic stellate cells (HSCs) in vitro .…”
