Novel Neuroprotective Function of Apical-Basal Polarity Gene Crumbs in Amyloid Beta 42 (Aβ42) Mediated Neurodegeneration

Abstract: Alzheimer's disease (AD, OMIM: 104300), a progressive neurodegenerative disorder with no cure to date, is caused by the generation of amyloid-beta-42 (Aβ42) aggregates that trigger neuronal cell death by unknown mechanism(s). We have developed a transgenic Drosophila eye model where misexpression of human Aβ42 results in AD-like neuropathology in the neural retina. We have identified an apical-basal polarity gene crumbs (crb) as a genetic modifier of Aβ42-mediated-neuropathology. Misexpression of Aβ42 caused u… Show more

“…Because of the recent discovery of the relationship between the crb and amyoid beta, it may be possible these three genes work together in the same genetic pathway (Steffensmeier et al, 2013). Our data strongly suggests that Tau plays important functions in the modulation of cell membrane domains including the apical Crb and AJs in photoreceptors during pupal eye morphogenesis.…”

“…Given the high degree of evolutionary conservation of crb and tau genes from Drosophila to higher mammals including humans, the similar cooperative mechanism between crb and tau could have a role in the development and degeneration of human photoreceptor. Because of the recent discovery of the relationship between the crb and amyoid beta, it may be possible these three genes work together in the same genetic pathway (Steffensmeier et al, 2013).…”

“…Adult Drosophila eye images were taken 70 using a Zeiss Apotome Imager.Z1 microscope (Carl Zeiss Microscopy GmbH, Jena, Germany). The flies were prepared by freezing them at −20 °C for ~2 h. The legs and wings of the flies were removed and flies were mounted on a dissection needle, and the fly was positioned on a glass slide using mounting putty.…”

“…Identified pathological changes in AD are an increased production of senile plaques, an accumulation of b-amyloid (Ab) protein generated by endoproteolytic cleavage of a 42 amino acid peptide with a high propensity of aggregation, reactive microglial cells, dystrophic neurites and bundles of astrocytic processes and intracellular neurofibrillary tangles 86,87 . The accumulation of b-amyloid protein leads to extensive neuronal degeneration and triggers neuronal cell death by unknown mechanism 88 . There is currently no cure for AD and, although available medications may temporarily ease symptoms, they do not slow the progression.…”
