Novel nail penetration enhancer containing vesicles “nPEVs” for treatment of onychomycosis

Abstract: Context: The systemic treatment of onychomycosis has been hampered by the reported side effects of antifungals in addition to the limited blood circulation to the affected nails. Topical ungual treatment would circumvent the limitations of systemic onychomycosis treatment. Objective: Preparation and characterization of nail penetration enhancer containing nanovesicles (nPEVs) loaded with sertaconazole for topical treatment of onychomycosis. Materials and methods: nPEVs were prepared using different nail penetr… Show more

“…They also increase the solubility of drugs, improve their bioavailability, provide a prolonged drug release and offer targeted drug delivery 1 . Penetration enhancer vesicles “PEVs”; which are composed of a penetration enhancer intermingled with phospholipids have recently presented themselves as a new versatile vesicular nanocarrier with wide variety of applications 2 7 ; namely for topical and transdermal delivery purposes. With the emergence of several functional materials that could be used in drug delivery, the term “nanocomposite” was introduced, which refers to a class of hybrid materials consisting of dissimilar components of different properties combined in one system 8 .…”

“…However, it still remains a great challenge due to the low permeability of the nail plate, recurrence, fungal resistance and consequently low cure rates (Bseiso et al, 2015;Flores et al, 2015;Piérard et al, 1993).…”

“…Owing to the surfactant and cosurfactant content of the microemulsion, it was reported to exhibit very small particle size. The negative charge exhibited by the microemulsion could be ascribed to the presence of ethanol and oleic acid [21,22]. Neither the particle size nor the charge was changed after 3 months storage, which indicated the stability of the microemulsion system.…”
