Novel methoxylated FA from the Caribbean sponge Spheciospongia cuspidifera

Abstract: The delta6 monoenoic methoxylated FA (6Z)-2-methoxy-6-heptadecenoic acid and (6Z)-2-methoxy-6-octadecenoic acid were identified for the first time in nature in the phospholipids from the uncommon Caribbean sponge Spheciospongia cuspidifera. These findings expand the occurrence of A6 2-methoxylated FA to C17-C18 chain lengths and establish a new FA biosynthetic possibility for these marine organisms. The novel methoxylated FA also could have originated from the phospholipids of a bacterium in symbiosis with the… Show more

“…Among the studied FA from P. suberitoides a novel amethoxylated fatty acid, namely 4, was also characterized using GC-MS as well as gas chromatographic ECL values as compared to synthetic standards [14,19]. The mass spectrum of the methyl ester of 4 displayed a molecular ion peak at m/z 286 and a strong M + -59 peak at m/z 227 (100%), together with a small peak at m/z 104 (McLafferty rearrangement), all typical mass fragments for a-methoxylated saturated methyl esters [14,19].…”

The a-methoxylation was further confirmed by 1 H NMR spectroscopy since it was also possible to observe the characteristic signals of the a-methoxy methyl ester in the 1 H NMR spectrum of the whole FAME mixture without interference from other peaks.…”

“…The final acid 2 was obtained after methylation of 13 with sodium hydride and methyl iodide in THF, following an acidic workup, resulting in the (6 Z )-(±)-2-methoxy-6-octadecenoic acid ( 2 ) in a 90 % yield. The overall yield for this eight-step synthetic sequence (starting from 5 ) was 13 %, and this is the first reported total synthesis for 2 [12]. …”

“…Was synthesized from the catalytic hydrogenation of 3 in hexane with H 2 and 10% Pd/C and the purity of the final product was confirmed by 13 C-NMR spectroscopy as well as comparison of the MS spectral data with the one previously reported [12]. …”
